Research Abstract

ネコの感染症としてよく知られているトキソプラズマ(Toxoplasma gondii)感染症は原虫感染症の一つで、世界中に感染者が存在する。治療法として、ロキシスロマイシン(マクロライド系抗生物質)やスルホンアミド、ピリメタミン等(抗寄生虫薬)が処方されるが、強い副作用によりその利用は制限される。副作用の少ない効果的な治療法の開発が急務となっている。
本年度はまず金ナノロッドとロキシスロマイシンの共有結合による修飾を試みた。ロキシスロマイシンのオキシム基をアミノ基に変換し、金ナノロッド表面に修飾したカルボキシル基にアミド結合で修飾した。しかし、修飾後、金ナノロッド表面が疎水性となるためと考えられるが、水中での安定性が極めて悪く、実際の治療実験に用いることのできるものは作製できなかった。一方、共有結合ではなく、疎水性相互作用による修飾法も昨年度から試みてきたが、やはり、修飾後の金ナノロッドの安定性に問題があり、改善が必要であった。リポソーム内に金ナノロッドとロキシスロマイシンを内包させる手法も試みたが、これも安定性に問題があった。ロキシストマイシン自体の親水性化や他のより親水的な薬剤を利用することにより、この問題は解決できると現在考えている。
一方、金ナノロッドの発熱効果を利用した経皮ワクチンシステムの開発も昨年度に引き続き進めた。今年度はその内容を「D.Pissuwan et al., A solid-in-oil dispersion of gold nanorods can enhance trandermal protein delivery and skin vaccination, Small, 7, 215-220 (2011)」として、国際的な学術雑誌に報告し、同時に国内特許(金ナノロッドを含有するS/O製剤、特願2010-136286)も出願した。
