Research Abstract

Mannosidase II (Man II) shows rapid redistribution into the endoplasmic reticulum (ER) followed by degeneration of the Golgi stack under the effect of brefeldin A (BFA) or during mitosis. The redistribution of Man II is interpreted as the disappearance of the Golgi apparatus itself or diffusive mixing of the Golgi apparatus and the ER membranes. However, ultrastructural observation indicates the membranous cluster to remain that is apparently distinctive from the ER. GM130, a Golgi matrix protein, is detectable in not only intact Golgi apparatus but also mitotic or BFA-induced Golgi clusters without redistribution into the ER. After washout of BFA or during telophase, reassembly of Man II on GM130-positive membranes and regeneration of Golgi stack can be observed. In postnatal development of rat parotid gland or under the effect of nocodazole, the primitive Golgi apparatus composed of small tubules and vesicles appears in the acinar cells, in which positive reaction for anti-GM130 and negative for anti-Man II can be recognized. The expression of Man II on the Golgi apparatus coincides with the establishment of a stacked structure at later developmental stage of the cell. From these results, we concluded that most of the membranous structure is the basic and plastic elements of Golgi apparatus on which GM130 is closely associated, and Man II containing membrane is the functional, additional and removable element. Integrated structure and normal function of the Golgi apparatus could be established after addition of functional elements to basic ones.
