Research Abstract |
It was confirmed that the BChE-positive fibers were distributed in inner and outer portions of rat frontal cortex using newly developed butyrylcholine esterase (BChE) histochemial method. This distribution is clearly different from those of the cholinergic fibers observed with the conventional histochemistry. The ascending BChE-positive fiber bundle was traced from sub-cortical areas followed the medial forebrain bundle and divided both into the medial and lateral sites by the basal ganglia then toward the frontal cortex. There are BChE-positive neuron groups, which project their fibers to the frontal cortex, consist of the pedunculopontine tegmental nucleus (PPTg) and the laterodorsal tegmental nucleus (LDTg) in the midbrain cholinergic system and noncholinergic nuclei in the thalamus such as the anterior nuclei of thalamus. d with B ChE histochemistry. Neuronal cells of the pedunculopontine nucleus and LDTg were injured by ibotenic acid(IA) for identifying the origin of B ChE-positive fibers in the frontal cortex. By an injection of FG in lateral frontal cortex, labeled large size neurons were observed in PPTg and LDTg. It was the 8-15 neurons (1 1 neurons average) in all sections. The labeled neurons were localized mostly in PPTg of the injection side. There was few neurons in the laterodorsal tegmental nucleus and the contra lateral of PPTg. Labeled neurons were abounding in the oral part ofPPTg and not observed in the caudal part. More than half of these neurons (60%) were BChE-positive. BChE-positive fibers in the outer part of the frontal cortex of the injury side has disappeared by the injury with IA to PPTg. On the other hand, there was no change on BChE-positive fibers of medial frontal cortex. It was confirmed that direct nerve fiber projected toward lateral side of the frontal cortex from the cholinergic neurons of PPTg.
