Outline of Research at the Start

本領域研究は、有機合成に破壊的イノベーションを起こすデジタル有機合成(実験科学と情報科学の異分野融合)の基盤を世界に先んじて築くことを目的とし、有機合成の多様性に対応した独自のデジタル化プラットフォーム(PF)を構築する。A01班、A02班、A03班の3班体制で、人工知能(AI)を徹底活用した自動化法(分子構造自動設計、合成経路自動探索、反応条件自動最適化、バッチ→フロー自動変換、自律的自動合成システム)でムダを徹底排除し、革新反応・革新分子創出の超加速化を実現するとともに、自動化法開発の基盤となる、有機化学の機械学習に最適化した本領域独自のデータベース(DB)の構築を行う。

Outline of Annual Research Achievements

本領域研究では、革新反応開発に機械学習を徹底活用(実験科学と情報科学の異分野融合)することで、化学選択性の触媒制御法開発の超加速を実現し、その基礎となる官能基標的触媒を網羅的に創出することを目的とし、(1)機械学習による反応条件最適化の超加速、(2)官能基評価キット活用による化学選択性の網羅的データ集積、(3)機械学習による新反応制御因子の顕在化、(4)機械学習によりデザインされた革新分子の合成と評価、(5)開発した革新反応の反応モジュール化とフロー反応での活用の検討を行っており、総括班はこれらの研究を円滑に遂行できるようにするために様々な取り組みを行っている。R5年度は主に以下の検討を行った。
(1)実験化学者が機械学習を取り入れながら研究を行うための研究環境整備を行なった。①電子実験ノート(SignalsNotebook)からAPIを利用して、領域データベース用のフォーマットでデータを抽出する方法を完成させ、約2万個のデータを集積した。②対面式の機械学習勉強会を2回実施した。これらの勉強会は録画しいつでも視聴可能となっている。③①と②の環境整備を基盤に、機械学習を活用した共同研究を推進した。
(2)化学選択性の網羅的情報収集のための官能基評価キットを大量に準備した。合成が必要な4つの化合物は当初受託合成を依頼していたが、フロー合成による供給体制を整えた。準備した評価キットを多くの班員に配布し様々な反応において評価を行った。
(3)(5)フロー合成装置を有する拠点の設備をさらに補強し、共同研究を推進した。
(4)2回の国際会議を主催した他、各種シンポジウムでの発表や共催などを通じ、領域の活動の広報を行い、さらなる産学官連携強化を行なった。

2: Research has progressed on the whole more than it was originally planned.

Reason

(1)まず、実験化学者向けに機械学習の研究環境整備を行った。各班員および学生に向けた教育動画を更に充実させるとともに、進展の大きい情報科学の知識と活用法をアップデートするために、新たに2回の演習形式の勉強会を行った。連続型変数のベイズ最適化の検討は全ての班員が研究に利用できるようになった。また、領域データベースのフォーマットに合わせたデータの抽出ができるように、電子実験ノート用にテンプレートの作成とAPIの修正、GUIの作成を行い、誰でも簡単にデータの抽出を行えるようにした。溶媒効果や触媒構造などのデジタル化が困難な離散型変数の最適化のために総括班で立ち上げたプロジェクトを推進した。特に、溶媒効果に関しては、最適溶媒を提案可能なシステムを数種類構築し、それぞれの有効性を検証する段階となっている。金属触媒や有機触媒に関する機械学習に必要な特徴量を網羅したデータベースの構築も継続して実施している。
(2)網羅的な官能基共存性の評価を行うために構築した官能基評価キットを用いて、実際に様々な反応の評価を実施する段階となっている。予想外の発見も多く、評価キットを利用したシステムの有用性が示されている。
(3)電解反応や光反応のフロー反応化を行うための共同研究を推進し、ベイズ最適化を用いた反応条件最適化の効率化に成功した。また総括班で新たなプロジェクトを立ち上げ、フロー化が実現できていない反応のフロー化を検討するシステムを構築した。
(4)R5年5月と12月にそれぞれ領域主催の国際会議を行い、領域に関わる最先端研究の発表と情報収集を行うとともに、参加者間で意見交換を行い、今後の国際共同研究推進をはかった。また、国内の主催および共催のシンポジウムを開催し、また多くの招待講演を行うことで、広報活動に努めるとともに、産官学連携を強めた。

Strategy for Future Research Activity

R6年度はこれまでの検討結果をもとに、(1)から(5)の項目に関して、以下の検討を行う。
(1)総括班として機械学習の教育用動画を更に充実させる。これまで実施していた機械学習をチュートリアル形式で学ぶ若手勉強会に加え、フロー反応や電解反応を学ぶ若手勉強会、遷移状態探索を効率化できるGRRMの講習会などを実施する。電子実験ノートから領域データベースへのデータ提供を簡便に行うワークフローの領域内での普及を進める。データベースの将来的な拡張を企図し、領域データベースへの領域外の研究者の参画を推進するとともに、構築したデータベースの公開の準備を進めていく。(2)官能基評価キットを活用した化学選択性の網羅的情報収集のワークフローに関しても、領域外の研究者に開放し、その利用促進を図る。また、得られたデータから化学選択性のデータベースを構築し、そこから官能基共存性を予測する機械学習手法の構築を検討する。(3)引き続き、代表的な不斉金属触媒、不斉有機触媒の特徴量を網羅したデータベースの拡充を図り、計算科学と情報科学を融合させる。(4)デジタル創薬に関する共同研究をさらに拡充する。構造活性相関研究の対象を創薬から機能性材料に広げ、幅広いケミカルスペースでの有用物質探索を推し進める。(5)バッチ反応のフロー反応化を推進するために、総括班で新たに立ち上げたプロジェクトを活用し、フロー化が実現できていない反応のフロー化をを推し進める。また、固相担持触媒反応、電解フロー反応、光フロー反応などの検討を加速するための共同研究体制をさらに強化する。
