Matrix-assisted laser desorption/ionization (MALDI) mass spectrometry is widely used in the analysis of biomolecules such as proteins. However, the mechanism of MALDI has not been fully understood. If the MALDI process is elucidated and controlled, further progress in the biomedical fields is expected. In this study, we have used a mid-infrared laser instead of a conventional ultraviolet laser for MALDI and have succeeded in ionizing an insoluble analyte which has been hard to ionize. In addition, we have found that the wavelength dependence of the ionization efficiency of peptides is correlated with the chemical structure of the matrix and that the water solvent has influence on the ionization with MALDI.
