Outline of Research at the Start

油体周期をオルガネラ周期のモデルとし，油体形成過程の高精細ライブイメージングと画像解析に一細胞トランスクリプトーム解析を組み合わせ，油体周期の各相を分離することで油体周期の実態を明らかにする．また，生体分子ダイナミクス解析により油体周期の中心振動子を推定するとともに，その分子機能を実験的に検証する．続いて環境変動を与えた際の油体周期の変調が油体の分布パターンの変化として表出する仕組みを解明する．

Outline of Annual Research Achievements

油体周期仮説では、細胞膜期と油体期が相互に転換することを想定している。その証明のために油体細胞一細胞におけるトランスクリプトームの変遷を調べることを目指しているが、核の単離条件の検討が難航している。現在他の植物でうまくいっている方法をゼニゴケに適用すべく、実験条件の検討を続けている。一方、油体形成のメカニズムの解明においては大きな進展があった。発生中の無性芽では、通常は一つの油体細胞あたり一つの巨大な油体が形成される。そこに微小管重合阻害剤を処理すると、油体が断片化することが見いだされた。アクチンの重合阻害剤は、このような影響は持たなかった。このことから、油体形成には微小管が深く関わることが示された。そこで蛍光タンパク質で標識した微小管により油体形成時の微小管のライブイメージングを行ったところ、形成中の油体から放射状に伸びた微小管構造が観察された。我々の研究から油体は細胞板と類似した性質を有していることが分かっている。そこでこの構造を油体フラグモプラスト（OB phragmoplast）と名付け、微小管の配向などの解析を進めている。続いて、油体形成のマスター制御因子であるERF13を条件的に発現する実験系を構築した。estradiol添加によりERF13の発現を誘導したところ、光合成細胞に分化した細胞を含むほぼ全ての細胞で油体が形成された。このことは、ERF13が細胞の分化状態を転換する能力を持つことを示唆している。さらに、ERF13を誘導すると、ほぼ全ての細胞でOB phragmoplastが形成されることも見いだした。現在はOB phragmoplastの構成タンパク質を明らかにするべく実験を進めている。
油体の形態が異常になる変異体の解析も進めた。油体形態が真球に近くなる変異体の原因遺伝子が被覆複合体であるCOPIの構成因子MpSEC28であることを突き止め、その細胞内局在などを詳細に明らかにした。

Strategy for Future Research Activity

引き続きバイオイメージング解析を中心とした油体周期仮説の検証と油体形成機構の解明を進めるとともに、単一油体細胞核トランスクリプトーム解析に向けた条件検討、油体形態が異常になる変異体の解析を進める。
