Outline of Research at the Start

超地球生命体の持続的成長を維持するためには、微生物の種と機能に裏付けされたポストコッホ機能生態系を理解することが重要である。アーキアは、未だ培養種の数は少なく、その生命機能に関する研究は立ち遅れている。本研究では、アーキアの種と代謝機能の多様性を解明することを目的とする。そのために、多様な環境中から未培養系統群を含むアーキアの単離あるいは集積培養を進め、それらを対象に（メタ）ゲノム解析を行い、配列情報に基づいてアーキアの代謝ポテンシャルを推定する。また増殖特性解析、生化学的解析などを通じて、アーキアの未同定代謝機能およびそれらを支える酵素・代謝経路とその制御機構を解明する。

Outline of Final Research Achievements

In addition to isolating novel extremophiles, we have identified enzymes from archaea that catalyze new biological reactions (e.g. arginine synthetase), display novel structures (e.g. dephospho-CoA kinase, lipoyl synthase) and new metabolic pathways (e.g. non-carboxylating pentose bisphosphate pathway, arginine synthetase pathway, archaeal glycolate pathway). These findings have contributed greatly to our understanding of metabolism in archaea.

Academic Significance and Societal Importance of the Research Achievements

アーキアにおける代謝に関して、基盤代謝の多くの不明点を明らかにした。Archaeal glycolate pathwayの発見により、光呼吸経路など2-phosphoglycolateの解毒機構が酸素発生型光合成システムやCBB回路が進化する以前から出現していた可能性を示唆し、従来の認識を覆すものである。またarginine synthetaseはアーキアから同定できたが、その分布はバクテリアや真核生物に及び、生物のアミノ酸代謝の理解に大きく貢献するものである。
