Outline of Research at the Start

超地球生命体の生態系を微生物の種と機能に基づいて理解する「ポストコッホ機能生態系」を構築するためには、土壌優占種にとどまらず、希少微生物種の機能の解明が必要である。本計画研究は、希少放線菌に対象を絞り、モデル圃場等から未分離の希少放線菌を分離するとともに、既知種を含めた希少放線菌の新たな生理機能を明らかにすることにより、微生物の種と生理的性質の多様性を解明することを目的とする。

Outline of Annual Research Achievements

(1) モデル圃場を含む分離源からの希少放線菌の分離
従来法に工夫を加えた分離法として、消毒剤による土壌の前処理を行い、107種を新たに分離した。従来法でも182種を分離した。このうち、70種はrDNA配列より新種であると考えられた。また、生育促進能を有する放線菌を共培養菌とし、マイクロデバイスを用いた共培養による分離を行った結果、共培養菌なしの条件では分離されなった3種を分離した。
(2) 希少放線菌の新たな生理機能の解明
16株の培養液をLC/UV/MS解析を用いたPCスクリーニングに供し、2株から化合物の単離・構造決定を行った結果、2種の新規化合物を同定した。ゲノムマイニング研究では、１個の遺伝子クラスターについて異種発現、代謝産物解析を行い、新たに生産される代謝産物を見出し構造解析に着手した。一方、昨年度解析した遺伝子クラスターの１つについて、2個の鍵酵素遺伝子だけを発現し、カフェ酸が生産されることを見出した。二次代謝の生合成研究では、スピナマイシン、p-クマル酸、アザセリン、シラチオマイシンの生合成経路について4報の論文を発表した。モデル希少放線菌Actinoplanes missouriensisの研究においては、胞子嚢胞子の酸化ストレス耐性や休眠と覚醒に関与するユニークなシグマ・アンチシグマ系、胞子嚢形成の初期段階に関与するSsgB、胞子嚢の生理的成熟に必要なBldC、胞子嚢形成に関与するアシル基転酵素、胞子嚢胞子の切り離しに関与するアミダーゼについて5報の論文を発表した。また、胞子嚢膜のトリアシルグリセロールを分解するリパーゼの存在を実証するとともに、胞子嚢マトリクス多糖の精製方法を確立するなどの重要な成果が得られた。Sporichthya属放線菌の２種の完全長ゲノム配列を決定し、形質転換系の開発に成功した。そのユニークな細胞増殖形態について新たな知見が得られた。
