Outline of Research at the Start

細胞が自身の成分をリソソーム／液胞に運び、分解する現象をオートファジーと呼ぶ。近年、オートファジーには、そのプロセスや分解対象に様々な多様性があることがわかってきた。その1つであるマクロオートファジーについては、大隅博士がノーベル賞を受賞したものの、まだ多くの謎が残されている。それらの解明は生命現象を理解する基礎研究としての重要性に加え、マクロオートファジーが関与する様々な生理機能や疾患の研究における重要な情報基盤となる。本研究では、マクロオートファジーのプロセスと分解対象選択のメカニズムを分子レベルで理解することを目的とする。

Outline of Annual Research Achievements

(1) マクロオートファジーにおける膜動態の解明：シード小胞（Atg9小胞）とAtgタンパク質が構成する複数の複合体の間の相互作用の解析を進め、PI3K複合体IのPAS局在化機構を明らかにした。成果を論文にまとめ、投稿し、査読者のコメントに応じて改訂し、改訂版を投稿した。一方で、Atg2脂質輸送タンパク質複合体の小胞体への結合を制御するメカニズムに関して突破口となる発見が得られた。
(2) マクロオートファジーにおける新たな基質選択機構の解明：Atg24複合体が隔離膜の開口端に局在化し、開口部を拡張することで、リボソームやプロテアソームなどの大きな細胞質粒子の隔離膜内への取り込みに重要な役割を果たしていることを明らかにした。成果を論文にまとめ、投稿し、査読者のコメントに対応するための追加実験をほぼ完了した。
(3) マクロオートファジーとミクロオートファジーの連携：マクロヌクレオファジーに必須のATG39を破壊した出芽酵母は窒素飢餓下で早期に死に至る。この細胞死の原因はミクロヌクレオファジーの異常昂進が原因であることとその詳細を明らにし、成果を発表すべく、論文の執筆を開始した。
(4) オートファジーによるタンパク質分解の総合的理解：窒素源飢餓条件においてオートファジーによって選択的に分解される新規タンパク質を複数発見し、オートファジーのマシナリーがそれらを選択するメカニズムを明らかにした。これらの成果を発表すべく、論文の執筆を開始した。

Reason

本研究は、次年度(2023年度)が最終年度となるが、上記の通り、複数の課題について、論文を発表あるいは執筆する段階に達している。一方で、セミインタクト細胞を用いたオートファゴソーム形成の試験管内再構成など、期待通りの進捗が得られなかった課題もあるため、「おおむね順調に進展している」という自己評価とした。

Strategy for Future Research Activity

上記の通り、本研究で得られた成果を基に、複数の論文を改訂、投稿、執筆する段階に至っている。これらの課題に注力し、最終年度中に可能な限り多くの成果を発表することを第一の目標とする。一方で、残された課題も含め、今後の発展に向けて引き続き研究を実施する。
