Outline of Research at the Start

以下の3 点について研究を進める.1) マルチタスク遂行能力の評価法の確立.マルチタスクとして認知負荷下での姿勢制御を想定する.健常者と患者でのマルチタスク遂行能力の評価方法を確立する.マルチタスクの設計,および課題中の生理反応の計測プロトコルを樹立する.2) マルチレイヤ脳活動-身体統合モデルの開発.(a)脳活動ダイナミクスモデル,(b)感覚・運動制御系モデル,(c)身体筋骨格モデルの適切な接続によるモデル構築を目指す.3) モデルの検証 1)のデータを2)のモデルに組み入れ,モデル検証を行う.これより「神経伝達物質を考慮した行動遂行則の変容メカニズムの数理モデル構築」を目指す.

Outline of Annual Research Achievements

網様体脊髄路と前庭脊髄路を模した制御を導入した立位姿勢制御モデル(制御パラメータとして,筋緊張パラメータとフィードバック制御パラメータを持つ)を提案している.この制御モデルで異常姿勢での立位を実現するため,パーキンソン病患者の異常姿勢の解析を行った.ここでは,以下のプロセスを経た.1) パーキンソン病患者の姿勢データをもとに,筋骨格モデルを立位させられる筋緊張パラメータを計算する;2) 計算した筋緊張パラメータを設定した上で,動揺が小さくなるようフィードバック制御パラメータおよび姿勢を調整する.これにより,パーキンソン病患者様の立位を実現する筋緊張パラメータを推定できた.また,「筋緊張の増加に対して,異常姿勢における動揺がそれ以外の姿勢と比較して小さくなっているため,異常姿勢を呈する」という仮説が支持された.
マルチタスク下の姿勢制御における神経伝達物質の役割の検証を行ってきている.二重課題時に対するドーパミンの影響を調べるため,パーキンソン病患者を対象に,ON状態とOFF状態の2つの状態で,課題成績と脳活動を検討した.随意動揺の基礎実験を遂行し,ステッピングを用いた患者実験を遂行した.課題は,計算課題による認知課題と,ステッピングによる運動課題からなる二重課題とした.また,パーキンソン病で併存する事がある疼痛が,姿勢・運動パフォーマンスの低下にどう影響しているのかを明らかにするために健常者に実験的に疼痛を与えた研究を行った.

Reason

網様体脊髄路と前庭脊髄路を模した制御を導入した立位姿勢制御モデル(制御パラメータとして,筋緊張パラメータとフィードバック制御パラメータを持つ)の提案,さらにマルチタスク下の姿勢制御における神経伝達物質の役割の検証という観点から,当初の目的を達成しているため.

Strategy for Future Research Activity

今後の研究推進のためには,以下の2点が考えられる.
1. パーキンソン病診断にも使われるDopamine transporter single-photon emission computed tomography(DAT-SPECT)の解析と,モデル制御パラメータの関係の検証.
2. マルチタスク下の姿勢制御における神経伝達物質の役割の検証をするための追加実験.マルチタスク問題の起源に関する姿勢・運動での実験系構築のための基礎研究の遂行.
