Outline of Research at the Start

受精により全能性を有する1細胞期胚が生み出されるが、その際にそれまでの親世代の遺伝子発現プログラムが刷新され、新たに新世代のプログラムがスタートする。このプログラムは、発生を順調に進行させるために厳密に調節されていることが考えられるが、その実態はほとんど明らかにされていない。そこで、受精後に全能性を獲得した1細胞期胚からスタートする発生プログラムの完全理解に向けて、その基盤となる遺伝子発現プログラムの調節機構を明らかにする。

Outline of Annual Research Achievements

受精後の遺伝子発現プログラムの進行は、1細胞期から2細胞期にかけての著しい発現パターンの変化から始まるが、その調節の一部にDuxが関与していることが近年報告された。しかしその後、Duxをノックアウトしても発生への影響が小さいことが報告された。そこで、Duxにその構造が類似したタンパク質がDuxの機能を相補していることを考え、その候補を探索したところDuxblが見つかり、そのｍRNAを1細胞期と2細胞期で調べたところ高いレベルでの発現が見られた。そこで、この時期の遺伝子発現におけるDuxblとDuxの遺伝子発現調節への関連を調べた。
まず、Duxblを過剰発現させた2細胞後期胚の遺伝子発現をRNAseqで解析したところ、Duxのターゲット遺伝子群の半数以上に顕著な発現上昇がみられた。さらに、Duxのターゲット遺伝子には、Duxの発現量が減少する2細胞後期に同様に発現が減少するものと、そのまま維持されるものがあることが分かった。そこで、これらの遺伝子発現へのDuxblの関与をその過剰発現胚で調べたところ、前者の遺伝子群にはDuxblの過剰発現は影響を及ぼさなかったが、後者のものについてはDuxblによる影響が見られた。また、DuxのcRNAを顕微注入することにより、本来Duxの発現が減少する2細胞後期においてもその発現を維持させたところ、その後の発生が抑制された。これらの結果より、2-cell後期以降の発現を維持させたいものについてはDuxblよりそれらの発現を維持し、一方で発現を低下させたいものはDuxblによって調節されずDuxの発現減少に伴って発現が低下しているのではないかと考えられる。このようにDuxblがDuxのターゲットの一部の遺伝子の発現を調節していることが、1細胞期から2細胞後期への遺伝子発現プログラムの進行に関与しているものと考えられた。
