Outline of Research at the Start

受精卵は胎児と胎盤の両方に分化可能な全能性を有するが、全能性の分子基盤は不明である。本研究では、全能性消失時に生じるエピゲノムの変化を同定し、それを操作することで一旦消失した全能性を再獲得する技術基盤の創出に挑戦する。さらに、全能性細胞だけが有する「受精卵から胚体外組織への発生系譜」に特徴的なエピゲノムの維持機構を解明する。

Outline of Annual Research Achievements

受精卵は、胚体組織と胚体外組織の両方に分化可能な潜在能力である全能性を有する。この全能性は、内部細胞塊(ICM)と栄養膜細胞(TE)への最初の分化が生じる胚盤胞期までに消失する。全能性の維持と消失には、エピゲノムの可塑性の維持と不可逆的な変化がそれぞれ重要と考えられるが、その分子実体は不明である。

本研究では、受精卵の父母アレルが有する機能の違いに着目して、全能性の理解に向けた研究を進めている。受精卵のエピゲノムの大きな特徴に、父母アレル間のヒストン修飾のゲノムワイドな非対称性がある。この父母アレル間非対称性は着床前発生過程で大きく失われるが、例外もあることがわかってきた。例えば、ポリコーム抑制複合体2 (PRC2)により付与されるH3K27me3修飾は、母性アレル特異的な局在を維持しながら発生し、DNAメチル化非依存的なゲノム刷り込みを制御する。そしてこの刷り込みの破綻は胚発生・胎盤形成の異常を引き起こす。このことから、配偶子形成過程におけるヒストン修飾の確立機構や受精後の動態の解明は、全能性の理解に重要であると考えられる。

本年度、井上は、卵形成過程におけるH3K27me3の確立機構、H3K27me3依存的刷り込みの機能、および、受精後に起こる発生関連遺伝子群からのH3K27me3の消失に関する研究を進めた。山口は、全能性に類似した性質を示す2細胞期胚様胚性幹細胞（2CLC）において、リボソームRNAプロセシング関連遺伝子の異常とp53の活性化がESCから2CLCへの移行を促進することを見出した。また、2細胞期胚の核の特徴であるクロモセンター形成に関する研究を発表した。

Reason

（井上）　胚発生・胎盤形成過程におけるH3K27me3依存的刷り込みの機能に必須な遺伝子座の同定に成功した。また、卵形成過程におけるH3K27me3の確立に関与する新たな因子を同定した。さらに、受精後のH3K27me3の消失に関して、その機構と意義を解明しつつある。（山口）　2CLCというin vitroモデルを利用し、p53を通じた全能性細胞の移行制御機構を発見し、報告した。さらにストレス応答が異なる多能性の移行に機能している証拠が得られており、その背後にある分子機構にアプローチする準備が整った。

Strategy for Future Research Activity

【井上 計画-1】H3K27me3依存的刷り込みの確立に、H2Aモノユビキチン化修飾を必要とする遺伝子群とそうでない遺伝子群があることを見出している。現在、このH2Aub依存性の違いを生み出す機構について解析を進めている。
【井上 計画-2】受精後の発生関連遺伝子群におけるH3K27me3消失の機構と意義に関して研究を進める。
【山口 計画-1】胚性幹細胞（ES細胞）中に存在する2細胞期胚様細胞（2CLC）への移行をモデルに、全能性の獲得、消失機構の解明に取り組む。これまでにp53が2CLCへの移行に重要な機能を果たしていることを報告したが、今年度はp53の上流、下流で働く機構として細胞周期と複製ストレスに着目して研究を展開する。
【山口 計画-2】 全能性消失期における父母アレルの働きの違いを明らかにする。これまでにキメラ胚を用いて特定の細胞系譜を単為発生胚由来細胞に置き換える系を確立した。今年度はこの系を用いて、内部細胞塊やその他の組織における父母アレル特有の機能や制御機構を明らかにする。
【山口 計画-3】DNA脱メチル化関連酵素であるTet1とPRC1の主要サブユニットであるRing1Bに機能的な相互作用があることを示唆する結果を得ている。そこで今年度はTet1欠損マウスおよびES細胞を用いて、この相互作用の機構と意義を詳細に解析する。
