Outline of Research at the Start

精子と卵子の融合により形成される受精卵や、体細胞核移植胚においては、大幅なエピゲノムの書き換え、すなわち「エピゲノム再プログラム化」を介して全能性が獲得されると考えられている。しかしながら、技術的問題から、その分子機構は未解明のままとなっている。本研究では、最新の技術の応用と新規技術の開発によって、その分子実体に迫り、全能期のエピゲノム制御機構を詳細に理解する。さらに、それらの結果を基盤として、全能性の人為的制御へと発展させる。これらの研究から得られる成果は、生命の始まりの基礎的理解にとどまらず、近年ますます増加傾向にある生殖補助医療にとっても重要な知見となることが期待される。

Outline of Annual Research Achievements

本研究では、受精卵に特有の全能性の理解のために、受精卵の有するエピゲノム状態およびそれと連動する転写状態を明らかにすることを重視した。そして、受精後発生における転写パターンの変化を正確に捉えるための手法の開発に取り組んできた。受精直後の胚では卵子由来の母性RNAが大量に存在するために、どのゲノム領域が実際に転写されているのかは明確になっておらず、全能性受精卵の転写パターンは不明であった。そこで、受精後に新規に合成されたRNA（nascent RNA）のみを網羅的に検出することのできる方法（LET-seq）を開発した。１００個のES細胞で質の高いデータを得ることができることを確認した後、受精後胚に適用することで異なる発生段階での転写状態を明らかにした。これにより受精後の転写ダイナミクスが明らかとなったとともに、全能期の転写制御因子としてObox3を同定した。全能性獲得が不十分なために発生異常を起こす体細胞核移植胚にObox3を導入すると、その発生が大幅に改善した。これらの結果は論文として発表した（Sakamoto et al. Cell Rep 2024)。また、受精後発生におけるクロマチン状態変化を明らかにするために、MNase-seqの微量化に取り組み、手法の開発に成功した。そして、マウス初期胚に適用することで受精後発生過程におけるヌクレオソームポジショニングの動態を明らかにすることができた。特に、受精直後の全能期のヌクレオソームポジショニングは、より発生のすすんだ胚や細胞のそれと比べ大きく異なること、転写因子YY1がヌクレオソームポジショニングの制御に重要な機能を果たすことを見出した。また、胚性ゲノム活性化とヌクレオソームポジショニング変化との関連性を明らかにした。これらの結果は論文として発表した（Sakamoto et al. Genes & Dev 2023)。
