Outline of Research at the Start

本研究では、遷移金属を構成要素とするセンサーモジュールを有したセンサータンパク質の生合成過程におけるセンサーモジュールの構築とタンパク質部分への組込みの詳細な分子機構を解明することにより、細胞内生命金属動態制御を鍵反応とするセンサー分子システムの構築原理を理解し、「生命金属科学研究基盤の構築」のための基礎的知見を得る。さらに、センサーモジュールによる気体分子センシングの動的過程、気体分子センシングにおいて誘起されるタンパク質構造変化・機能制御を各種分光学的測定、結晶構造解析、生化学・分子生物学的解析によって明らかにし、生命金属が制御する細胞内構造ダイナミクスと機能の解明を行う。

Outline of Annual Research Achievements

本研究では、HemATによる酸素センシング、および酸素をシグナル分子とする走化性制御系におけるシグナル伝達の分子機構を解明するため、HemATセンサードメインについて酸素化型と還元型の結晶構造をそれぞれ2.5Å 、2.36Å分解能で得ることに成功した。各状態を比較したところ、酸素がヘムに配位することで、ヘムが配位しているHis119が回転し、回転に伴って隣接するTyr129の側鎖の角度が大きく変化していることが確認された。またシグナルON（酸素化型）では、Tyr129の変化に伴って、Tyr129とシグナリングドメインに続くヘリックス内に位置しているTyr168間で水を介した水素結合ネットワークが新たに形成されている一方、シグナルOFF（還元型）ではその水素結合が形成されていないことを確認した。さらに、シグナリングドメインに続くC末端ヘリックスについて、酸素化型ではC末端ヘリックスの電子密度が見えているのに対して、還元型では見えていない領域があることが確認された。これは、還元型のC末端ヘリックスが不安定であり、各状態で構造安定性に差があるためであると考えられる。これらの結果から、ヘムに酸素が配位することによってC末端ヘリックスへと続く水素結合が形成され、構造が安定化される一方で、酸素が配位していない場合では水素結合が形成されないことでC末端の構造が不安定化され、この構造安定化の違いがシグナリングドメインに対するシグナリングとして伝達されていることが推察される。
さらに本研究では、HemAT/CheA/CheW複合体について、クライオ電子顕微鏡を用いた単粒子解析を行ない、低分解能マップを得ることに成功した。複合体について得られたマップをもとに複合体構成比を検討したところHemAT：CheA：CheW＝2：1：1で複合体を形成していることが示唆された。

Reason

本研究では、ヘムを酸素センサーモジュールとして利用している酸素センサータンパク質HemATの構造機能相関解明、および酸素センシングならびにシグナル伝達反応の中心的な役割を果たすHemAT/CheA/CheW複合体による走化性制御の分子機構解明を目的とし、HemATならびにHemAT/CheA/CheW複合体の構造決定に向けて、X線結晶構造解析およびクライオ電子顕微鏡による単粒子構造解析を併用して研究を進めている。HemATの構造解析においては、シグナルON型およびシグナルOFF型の状態にあるHemATセンサードメインの構造決定に成功している。全長型HemATについては、いくつかの条件で結晶を得ることはできているが、まだ構造決定には至っていない。クライオ電子顕微鏡単粒子構造解析によるHemAT/CheA/CheW複合体の構造決定においては、低分解能でのマップを得ることが出来ており、現在、その分解能向上を目指して各種実験条件の検討を行っているところである。

Strategy for Future Research Activity

今後の研究では、酸素に対する走化性（Aerotaxis）制御系の構造機能相関解明を目的として、走化性制御系を構成する個々のタンパク質、および走化性制御において中心的な役割を果たすタンパク質複合体（HemAT/CheA/CheW複合体）の構造解析に向けて、X線結晶構造解析およびクライオ電子顕微鏡による単粒子構造解析を併用して研究を進める。これまでの研究において、HemATセンサードメイン、CheWタンパク質のX線結晶構造解析に成功しているので、今後は全長型HemAT、CheA、およびHemAT/CheA/CheW複合体の結晶化条件の検討・最適化を進める。
また、クライオ電子顕微鏡単粒子構造解析によるHemAT/CheA/CheW複合体の構造解析を行い、分解能7.36オングストロームでのクライオEM構造を得ることに成功しているので、今後はより高分解能での構造解析を目指し、各種実験条件の検討を進める。
また、水素センサータンパク質生合成反応の分子機構解明を目的とした研究にも取り組む。今後は、クライオ電子顕微鏡による単粒子構造解析を用いた、水素センサータンパク質生合成に関与する一連のアクセサリータンパク質複合体の構造解析も計画している。
