Outline of Research at the Start

金属が関わる生命現象を分子レベルで理解するためには、金属イオンの代謝プロセスや輸送に関わる「金属関連タンパク質」の網羅解析が必要不可欠である。本計画研究では、我々が近年独自に開発したコンディショナルプロテオミクスの原理を応用して、金属関連タンパク質の細胞内動態を分子レベルで解明するための方法論を開拓する。具体的には、組織切片や動物/植物個体内で適用可能な金属応答性タンパク質修飾分子（ラベル化剤）を開発し、生体内に豊富に存在する亜鉛、銅、鉄をはじめ、あらゆる生命金属の生体内動態解析を実現する新手法の確立を目指す。

Outline of Annual Research Achievements

我々は、独自に開発したタンパク質解析技術であるコンディショナルプロテオミクスの原理を応用して、金属関連タンパク質の細胞内動態を分子レベルで解明するための方法論を開拓している。銅イオン（Cu+/Cu2+）に応答して反応性が上昇する新規ラベル化剤として、昨年度に引き続きキレーター部位を有する銅イオンコンディショナルプロテオミクスプローブの開発を行った。先行してChristopher Changグループ（UC Berkeley）と共同でアシルイミダゾール化学を基盤とする銅イオン応答性タンパク質修飾プローブを開発したが（J. Am. Chem. Soc.2020）、このプローブはCu+/Cu2+に対する選択性が低く、銅イオン非存在下におけるバックグラウンドも高いという課題があった。そこでさらなるプローブ性能の改善を目指して銅イオンとの錯形成に伴って高反応種であるキノンメチドを生成可能なプローブを設計した。その結果、新たに開発したプローブはCu+イオンに対して選択的であり、従来のプローブよりも低濃度のCu+によって応答することがわかった。この新規プローブを銅輸送タンパク質CTR1を発現したHeLa細胞に処置したところ、CTR1発現細胞選択的にCu+イオン依存的なタンパク質ラベリング反応が進行した。さらに、銅輸送タンパク質であるATP7Aの遺伝子ノックアウト細胞と野生型細胞を用いて銅イオンコンディショナルタンパク質ラベリングを実施した。その結果、ATP7Aノックアウト細胞では銅の細胞外排出が阻害されるため、野生型細胞と比較してよりラベル化反応が進行することが明らかとなった。さらに、ラベル化タンパク質を濃縮・精製し、LC-MSMSによる同定を行なったところ、いくつかの銅関連タンパク質が修飾されることが明らかとなった。

Reason

コンディショナルプロテオミクスに関するプロジェクトについては当初計画通りに進行しており、本領域最終年度となる2023年度には論文化を目指す。また、当初計画になかった成果として、我々が以前開発したオルガネラ選択的ホスファチジルコリン（PC）蛍光標識法をCRISPR-knockoutスクリーニングと組み合わせ、PCの細胞内存在量や局在といった表現型に摂動を与える遺伝子群を網羅的に同定する系の確立に成功した。O-ClickFCと名付けた本手法を用いた遺伝子スクリーニングの結果、予想外にヘムトランスポーターとして知られるFLVCR1がPCの生合成に関与していることが明らかとなった。さらなる解析の結果、FLVCR1は白血病細胞株K562細胞やその他様々な細胞種において主要なコリントランスポーターであることを発見した。FLVCR1はヒトの寿命に関与することが知られており、またその変異は様々な遺伝性疾患の原因となることが知られている。我々はPCARPやHSANといった遺伝性疾患で頻繁に見られるFLVCR1変異を持つ細胞ではコリンの細胞内取り込みが顕著に減少することを見出した。このことから、FLVCR1を原因遺伝子として有する遺伝性疾患の病態はコリン代謝異常によって引き起こされている可能性が本研究によって初めて示唆された。この成果は、2022年度にCell姉妹誌であるCell Metabolismに受理された。

Strategy for Future Research Activity

2023年度には銅イオンコンディショナルプロテオミクスプローブに関して細胞内での検証を進め、その成果を論文として発表する。また、O-ClickFC法については他の様々な代謝物や脂質への展開を目指す。
