Outline of Research at the Start

光によって感応・応答するタンパク質には、動物や植物などに広く分布する発色団を含み、生命維持に関与するなど重要な役割を果たすものが知られる。近年では、こうした光感受性タンパク質を利用して、細胞などの光操作する技術に応用され（光遺伝学）、これまでに多くの生理現象の解明に利用されてきた。本研究計画では、疾病治療への利用が期待されている、またはツールとしての重要性が高いとされる光感受性タンパク質について、XFELを用いた高速分子動画実験を行うことで、タンパク質内部で起こる光エネルギー移動や高速な構造変化について明らかにする。

Outline of Annual Research Achievements

光感受性アデニル酸シクラーゼ（OaPAC）については単結晶から得られた静的な構造がすでに明らかであったので、まずこの結晶化条件を微小結晶調製用に改良することを試みた。その結果、暗状態でのタンパク質の発現や精製を行うことで、改善を行いことができまた。また、回折能がある結晶化どうかは見た目では判断できないため、単結晶のX線結晶構造解析で用いる大きさの結晶の回折能チェックを実験室系X線回折装置で実施し、それを微小結晶調製にフィードバックすることで品質管理を随時行いながら微小結晶の大量調製方法の検討を行なった。
さらに、XFELを使用して、微結晶から2.4 Aの高解像度構造データを取得した。Swiss Light SourceでのAcoustic Levitation Diffractometer（ALD）を用いた常温でのX線回折実験では、超音波領域内に浮揚したフィルム上の結晶に対してX線を照射し、様々な方位からの反射点を収集することで、励起光照射から14秒後まで2秒ごとの時分割でのデータ収集を行い、2.2-2.4 Aの分解能で明らかにした。また、PF（Photon Factory）でのクライオ条件下での構造解析では、室温にて青色LEDで照射後、瞬時にクライオ吹付装置で凍らせる方法を用いて照射後3~15秒の状態の結晶構造を1.7 -1.9 Aの分解能で明らかにし、BLUFドメイン内のFlavinと近傍残基Gln48が近づく動きを観測し、その動きが3秒後にはGln48の動きが完了していることを示した。
