Outline of Research at the Start

生体分子モーターのin vitro での1分子計測を契機に、少数分子の非平衡系に対する統計力学・情報熱力学理論が近年大きく発展した。しかし、より生理的な条件あるいは細胞内での生体分子の挙動の理解には至っていない。本研究では、in vitro および細胞内での1分子計測と理論研究のフィードバックにより、細胞内のような本質的に非平衡な混雑環境を前提とした統計力学・情報熱力学の理論構築を行い、実際の生体分子機械に対してこれを適用することで、生体分子機械の設計原理の解明を行う。

Outline of Annual Research Achievements

本研究では、1) 生体分子モーターの多様性と設計原理、2) 細胞内非平衡環境の計測、3) 非平衡多体現象の基礎理論、の3つの項目について研究を行う。
このうち 1) 生体分子モーターの多様性と設計原理については、トレードオフ関係式の理論を発展させるとともに、これに対応した計測系を実現するための装置開発を進めた。一分子計測で測定可能なパラメータを用いて分子モーターの性能にかかわるトレードオフ関係式に対応した一分子計測実験を行うために、高い空間分解能と時間分解能を両立する構造化照明一分子顕微鏡の構築を進め、ナノメートル精度を達成するなどの予備的な結果が得られた。さらに、1分子計測実験結果に熱力学的不確定性関係を用いてエントロピー生成を推定する手法について、実験結果を模擬したシミュレーションで有効性が確認できた。
2) 細胞内非平衡環境の計測については、並進拡散と回転拡散の同時計測を行うための6次元高速一粒子追跡顕微鏡の開発を進めた。この顕微鏡とマイクロ流体デバイスを組み合わせる実験系を構築した。また、細胞内で拡散粒子に力学的摂動を与えるための操作系の開発も進め、in vitroの系で多数の微粒子を一様に操作できることを確認した。新たに、細胞内での拡散計測のために、自己組織化蛍光ナノ粒子を細胞内に発現させる実験系を確立し、生細胞内での並進拡散計測を再現よく実施することで、細胞状態と細胞質の流動性の定量的解析が進展した。
3) 非平衡多体現象については、クロマチン構造転移が1次転移となり遺伝子スイッチとして機能する条件の理論を踏まえ、多色全反射顕微鏡を用いた再構成クロマチンの計測がスタートした。

前年度までのコロナ禍による遅れを取り戻し、概ね計画通りに進展できている。

Strategy for Future Research Activity

コロナ禍による影響での遅延の影響が当初懸念されたが、その後の研究の進展により、遅延の影響は概ね取り返すことができている。しかし、半導体不足などの影響で機器類の納期遅延が多々見られるので、できるだけ前倒しに発注するなど納期遅延の影響を最小限に抑えたい。
