Outline of Final Research Achievements |
In this study, we focused on the bacterial repellent response and measured the localization of CheB to the cell poles, which is necessary for the adaptive phenomenon. We simultaneously measured the change in fluorescence intensity at the poles and the motor rotation after the administration of repellent substances, and estimated the dependence on the concentration of repellent substances, the correlation between the rotation and the localization, and the number of localized molecules. Furthermore, the response of mutants in which the CheB binding site of the receptor was deleted was also measured. In addition, we developed a system to simultaneously measure and analyze the responses of multiple bacteria to eliminate the variation among experiments. As a result, the variability was clearly reduced compared to individual measurements. We are currently investigating whether this remaining variability is unique to E. coli or to the experimental conditions.
