Outline of Research at the Start

大腸菌走化性システムは、外界の誘因・忌避物質が細胞極に存在する受容体に結合し、その情報は細胞内を通して各モーターに伝わり、モーターの回転方向が転換される。申請者は、走化性刺激受容において、協同性、適応性、自発性の3つの特徴が、受容体のメチル化レベルの変動という共通の分子機構で実現されている、という従来説とは全く異なる新しいモデルを提唱している。そこで、１細胞レベルでの走化性動態を、活性を保ったまま明らかにする。分子レベルの反応ネットワークと細胞レベルの応答をつなぐ格好のモデル系であり、領域内の理論グループとの共同研究体制を構築することにより実験と理論との有機的な融合を目指す。

Outline of Annual Research Achievements

本研究は，バクテリア走化性に関わるすべてのタンパク質の挙動を1 細胞レベルでイメージングすることにある．
特に， 走化性に関わるCheYとCheZ双方に蛍光色素を導入しFRET計測を行った．その結果，セリンなどの誘因物質投与後にFRET効率が下がり，数十秒後にFRET効率が復活した．これは適応現象を観察したことになる．今まで使ってきた大腸菌は複数の受容体が混在しているWildタイプを用いてきた．しかし，セリンに応答する受容体は，Tsrという受容体のみである．この受容体の混在とセリン応答に対する感度との関係を調べた．その結果，予想とは逆にTsr単体ではセリン感受性が減少した．これは受容体ファミリーが混在した状況の方が，セリンを含めてより感度が上昇するということになる．現在，Tsr単体にあえて他の受容体を混在させることでその応答を調べている．
また，適応現象に必要なCheBに蛍光標識を行い，細胞極への局在状況を計測した．イソロイシンなどの忌避物質投与後の極の蛍光強度，持続時間は，同じ条件においても実験結果がかなりばらつくことがわかり，その原因について検討した．その対策として，個々の大腸菌の応答を調べるのではなく，系全体の複数の菌の応答を同時に計測することによって実験誤差を減少させたときの応答を計測した．この系では蛍光とモーターの回転を同時に計測することはできないが，100個単位の大腸菌の応答を同時に計測・解析することが可能となる．その結果，個々の計測に比べて明らかにばらつきが減少した．このことは，実験の環境，季節，気温，湿度，研究者の体調によってデータがばらついていたことをしめす．しかし，まだ有意なばらつきは存在する．現在，このばらつきが大腸菌固有のものなのか，実験条件（菌の体積，長さ，濃度などなど）なのかを検討しているところである．
