Outline of Research at the Start

高密度の細菌集団に対して、均一かつ制御された環境下での計測を実現する独自デバイス「広域マイクロ灌流系」を拡張、活用して、細菌集団の協同的現象を記述する熱統計力学原理の理解と探求を目指す。特に対象とするのは高密度細菌集団の凝集現象であり、実験と理論の比較を通して、自己駆動粒子等の系に一般化された熱力学アプローチの有効性を検証する。また、以上の知見と、光トラップやフィードバック制御などの実験技術を組み合わせて、細菌の凝集状態の実験的制御を目指すとともに、細菌集団を記述しうる熱統計力学量の計測や概念の検証を行う。

Outline of Annual Research Achievements

本年度も引き続き、高密度細菌集団のモデル実験系の研究、特に(i)高密度細菌集団の非平衡相の研究と(ii)高密度細菌集団の支配因子の探求を行い、細菌集団の情報物理学の基盤づくりを推進した。また、課題期間終了を見据え、(iii)細菌集団において情報物理学を推進する新たなモデル系の開拓にも着手した。
(i)については、前年度までの研究で発見した運動性大腸菌集団のガラス化について、マクロ統計量の解析を一通り実施した。従来物質のガラス転移と比較し、その頑健性に加えて、棒状の細胞形状や自己駆動に由来するとみられるアクティブガラスの新奇な特徴も見出した。以上の成果は論文プレプリント・学会等で発表した。さらに、海外グループとの共同研究によって一細胞トラッキングを実現し、マクロな統計的性質と一細胞の運動の関係を調査した。また、培養条件や菌株の違いにより、相分離様の混雑化やネマチック配向秩序の発達など、異なる混雑化現象が出現することを見出した。
(ii)については、昨年度に引き続き、非運動性の大腸菌集団におけるコロニーの３次元構造とトポロジカル欠陥の関係を調査し、成果を論文発表した。また、バイオフィルム形成において遺伝子発現の不均一性を生む物理因子の探求を目指し、細胞外基質の重要構成物の発現量が、配向秩序や細胞分裂といった外的な特徴と相関する可能性を調査した。
(iii)については、細菌の集団運動を制御する手段として、基板上に設けられた構造物の形状効果に注目した。特に、擬二次元的な系における柱と細菌細胞の相互作用に着目し、実験と数値計算によってその特徴を明らかにするとともに、流体力学的な考察を行った。以上の成果は、論文・学会等で発表した。さらに、外的制御可能な細菌集団の例として磁気走性細菌にも注目し、観察系を立ち上げるとともに、磁場応答や酸素勾配応答に関する基本的な性質を調査した。

Reason

高密度大腸菌集団のガラス転移についての研究は本年度も順調に進展し、論文プレプリントとして成果公開に至ったほか、相分離様の混雑化が出現する実験条件の絞り込み、ネマチック配向秩序が関わるガラス転移など、次なる展開に繋がる新たな観察結果も様々に得ることができた。また、バイオフィルム形成時の遺伝子発現の不均一性が、配向秩序や細胞分裂といった物理的事象と関連する可能性を期せずして見出すことができ、その検証にも注力した。これは高密度細菌集団が示す物理的特徴と生命現象のつながりを調査するうえで重要なモデルケースと考えられる。以上の、想定以上の研究成果の確立に注力するため、光トラップ系の実験については当初計画より規模を縮小して取り組んだ。また、細菌集団において情報物理学を推進する新たなモデル系の開拓も順調である。以上を総合して、「おおむね順調に進展している」と考えている。

Strategy for Future Research Activity

本年度までの研究により、高密度の運動性大腸菌集団の混雑化には、配向ドメインを伴うガラス化、ネマチック配向秩序を伴うガラス化、相分離様の混雑化の少なくとも３パターンがあることが明らかとなり、それぞれが現れる実験条件の理解も進んだ。配向ドメインにを伴うガラス化については、本年度までの研究で物理的特徴が解明されたので、それと比較する形で残り２つの混雑化についても統計物理的な特徴づけを進める。また、配向ドメインを伴うガラス化についても、本年度までのマクロ統計量の解析に加えて、一細胞の動力学データも取得できたため、対相関関数や配置エントロピー等、熱力学的観点から重要な量の解析を進める。以上により、運動性の高密度大腸菌集団における非平衡ガラス相について、熱統計力学的な理解の構築を試みる。
光トラップ系については、本年度までの研究によって、高密度大腸菌集団のタイムラプス観察と併用が可能な実験系が準備できている。菌の捕捉の強さや遊泳状態への影響の評価を続けるとともに、ガラス転移や相分離様状態の出現に近い高密度細菌集団に対して菌体トラップをすることにより、ガラス状態や相分離状態の核生成を試み、それによって準安定状態も含めた相図の作成を目指す。
高密度細菌集団の支配因子の探求については、本年度までの研究によって、トポロジカル欠陥と細菌集団構造の関係を見出すことができた。今後は、デバイス形状や液晶配向制御等の手段を用い、配向秩序を制御することで細菌集団に影響を与える可能性を検討したい。また、配向秩序や細胞分裂といった物理的事象と遺伝子発現の関係の検証を進め、メカノトランスダクションなどの観点から機序の理解を目指す。このような取り組みを通して、高密度細菌集団という非平衡大自由度系の時空間動力学がトポロジカル欠陥などの少数因子でどこまで規定され、記述可能なのかという問に近づいていく。
