Outline of Research at the Start

時分割シリアルフェムト秒結晶構造解析(SFX)法による「高速分子動画」は、原子分解能で数10フェムト秒からミリ秒までの構造変化を捉える手法である。一方でSFXのデータは、多数のタンパク質分子の平均構造であり、また、電子状態などの詳細を知ることは困難である。本研究では量子古典混合計算による複雑な化学反応解析や、高精度力場を用いた大規模長時間シミュレーション、実験データとシミュレーションを融合させるハイブリッドアプローチなどの多角的な理論研究アプローチを活用することにより、反応の時間・空間・エネルギーに関する詳細な情報を得ることで、タンパク質の動的構造変化とその化学反応機構を解き明かす。

Outline of Final Research Achievements

In this study, we used a variety of theoretical and computational methods to elucidate the dynamic conformational changes of proteins and their chemical reaction mechanisms. Our approaches included chemical reaction analysis using QM/MM (quantum mechanics/molecular mechanics) calculations, large-scale long-time simulations with highly accurate force fields, and hybrid techniques that integrate experimental data with simulations. By incorporating data from molecular movie experiments, which capture structural changes at atomic resolution, we have gained a better understanding of these processes.

Academic Significance and Societal Importance of the Research Achievements

高速分子動画法を用いた実験データと、量子古典混合計算や分子動力学シミュレーションなどの理論・計算手法を組み合わせることで、タンパク質の動的挙動や反応メカニズムを詳細に解析することが可能になった。このような融合的手法は化学や生物物理学などの学術分野においての基礎理論の発展に貢献する。また、タンパク質の動的構造変化や化学反応機構を解き明かすことは、疾患の原因究明や新しい医薬品の開発、既存薬品の改良につながるものである。
