Outline of Research at the Start

マテリアルが最初に接触する生体要素、すなわちマテリアルと生体とのインターフェースである細胞膜を、高速ライブセル原子間力顕微鏡を用いて「まるごと」観察すると同時に、そこに関与する生体因子や相互作用により生じる細胞応答を蛍光イメージングする。これらの相関イメージングにより、「弱い相互作用」を分子・細胞レベルで定義づけ、生体がマテリアルと遭遇した時に呈する細胞動態を可視化する。これによって相互作用の強弱が、「拒絶」「ステルス（回避）」「寛容」のいずれを導くかのプロセスをデコードし、本領域の命題である「マテリアル・シンバイオシスとは何か？」を定義し、「物質共生」の理解に貢献する。

Outline of Annual Research Achievements

本研究で実施する、細胞表面で生じるマテリアルと細胞表面タンパク質の「弱い相互作用」が生じている現場である細胞膜の動態観察を実現するために必要な、高速ライブセル原子間力顕微鏡（atomic force microscopy，AFM；以下、高速ライブセルAFM）および蛍光顕微鏡による相関イメージング（correlative light and acomic force microscopy，CLAM）の実装を行ってきた。また、本研究グループは、研究領域内の測定拠点としての活動も求められている。本年度は、昨年度から継続してその体制整備を実施するとおもに、活用した研究を進展させてきた。
設置した測定設備は以下の通り。高速AFM2機（うち1台は共焦点レーザー顕微鏡とのCLAM用、他の一台は薄層斜光照明法（highly inclined and laminated optical sheet microscopy，HILO）とのCLAM用）、高速ディスク式共焦点顕微鏡3機（うち2機は2波長同時刻画像取得可能、2機は超解像ユニット実装）、全反射顕微鏡1機、広視野顕微鏡1機である。
また、高速ライブセルAFMの目標諸元（範囲:6.0 μm × 4.5 μm ×1.0 μm、xyzすべての方向に対して10 nmの等方性空間分解能、0.5 フレーム/秒（2秒に1枚の撮像）の時間分解能という基本性能）を安定維持できるようになった。
これらの機器を活用した共同研究実績は、領域内外で20件に及んだ。また、領域共催でこれらの機器を活用し研究のさらなる進展を目指したイメージング講習会を開催した。

Strategy for Future Research Activity

さらに超解像ユニットを一式導入し、さらなる観察の高解像度化が進める。イメージング拠点は従前どおり円滑な運営体制も維持する。 また、次年度以降、これらの機器を活用し、相互作用様式の定量解析や物質あるいは各種病原体等と細胞との相互作用と細胞応答解析をさらに進める。おって、さらなる共同研究の活性化につなげる。
