Outline of Research at the Start

脳内の複数の機能要素（神経回路、グリア、血管など）の統合メカニズムを探索し、末梢臓器や全身レベルでの応答と関連付けることは重要な課題である。本計画研究では脳機能の要となる神経回路の形成・成熟におけるグリア細胞と神経回路のダイナミックな応答を先端的なイメージング技術を適用し、個体レベルで解析することを目指す。個体の内部環境に応答して脳内の特定の領域の機能調節が起こり、その結果として脳からの情報が末梢にフィードバックされる、という仮説について、特に精神神経疾患のモデルに着目して研究を推進する。

Outline of Annual Research Achievements

本年度は以下の二項目について研究を実施した。
（１）脳細胞移植と二光子顕微鏡イメージングの統合によるグリアデコーディング：未分化な神経幹細胞を別の動物の脳内に移植することで細胞の分化成熟を誘導することが可能である。この細胞移植技術を個体レベルでのイメージング手法である二光子顕微鏡技術と組み合わせることが出来れば、神経細胞、グリア細胞それぞれに由来するシグナルの切り分けや細胞自律的な機能調節の存在の有無を明確に示すことが出来る。本年度はアストログリアを貪食機能に関連する分子をノックアウトした動物より培養し、増殖させた後に野生型マウスの大脳皮質に移植する実験を実施した。移植された細胞は大脳皮質の深部に生着し、野生型の細胞と同様の形態を生後に発達させた。この移植されたアストログリアと、内在性のアストログリアの境界部を同定して、この境界部を横切る大脳皮質錐体細胞の樹状突起に形成されるスパインの密度を定量し、境界部においてスパイン密度が有意に変化することを見出した。この実験結果はアストロサイトの持つ貪食機能が局所的にスパイン密度を制御しうることを示しており、グリアと神経回路の間に存在する精密な相互作用を示す例と考えられる。
（２）脳深部での神経細胞、上衣細胞、タニサイト、グリア細胞の相互作用：前年度までの研究により、大脳皮質より深い層に存在する海馬歯状回の神経前駆細胞とその除去に働くミクログリアの動態を個体レベルで観察するための手術方法を確立した。この手法を用いて個体レベルでのミクログリア機能を解析した論文を公表した。またexpansion microscopyによって組織を拡大し、実質的な解像度を上げてグリア細胞の微細な構造を解析する手法を視床下部の脳室近傍の組織に適用し、脳室表面に存在する新規の軸索様構造とグリア細胞の形態学的な相互作用に関するデータを取得した。

Reason

四年目の研究の進捗として、研究項目（１）に関連してはアストログリアのシナプスに及ぼす局所作用について明確な実験的証拠を得ることができ、研究項目（２）については海馬歯状回における神経前駆細胞の除去におけるミクログリアの役割を個体レベルで解析する研究について論文発表することが出来た。従って研究は順調に進捗していると考えている。

Strategy for Future Research Activity

本計画研究では脳機能の要となる神経回路の形成・成熟におけるグリア細胞と神経回路のダイナミックな応答を先端的なイメージング技術を適用し、個体レベルで解析することを目指している。その為には脳移植技術と個体レベルでのシナプスやカルシウムのイメージング技術の統合が最も必要とされる技術である。次年度には貪食機能が低下したアストログリアが近傍に存在することにより、神経回路の機能をどのように障害するのか、より機能的な解析を行うことで詳細を明らかにする予定である。また脳深部におけるグリア細胞の機能については、海馬歯状回と視床下部における解析を継続し、グリアによる回路の機能制御についての新知見を得ることを目指す。
