Outline of Research at the Start

本計画研究はミクログリアの置換技術開発により、ミクログリアデコーディングが全身監視・制御に果たす役割を、睡眠覚醒サイクル研究（史班）及びエクソソーム研究（星野班）との緊密な相互連関により明らかにする。先ず全脳レベルでミクログリアを非侵襲的に細胞置換する技術「リセット」及び「グラフト」を開発する。次いで種々ミクログリア置換が脳及び末梢機能に果たす役割を、神経及びグリア細胞の微細同時Ca2+イメージング法を主体として明らかにする。さらに、疾患iPS細胞由来ミクログリアの移植に挑戦し、各班に提供することでミクログリアデコーディングが媒介する脳及び末梢疾患を理解するチャレンジングな課題も行う。

Outline of Annual Research Achievements

ミクログリアは脳内及び全身機能を監視し、脳及び全身の恒常性を制御するインターフェースとして重要な役割を果たしている。本研究では、ミクログリアを置換する2つの技術を開発し、それらを用いてミクログリアの脳及び全身の機能をどのようにデコーディングし、脳及び末梢臓器の制御を行うのかについての検討を行うものである。ミクログリアの置換には、colony stiumlating factor1受容体（CSF1R）拮抗薬のON/OFFでミクログリアを除去/自己増殖により行う「リセット」及びリセットとミクログリア移植を組み合わせた「グラフト」により行った。一昨年度までにこれらのミクログリア置換法の開発が終了したので、本年は昨年に引き続いてミクログリアが脳内の異常をどのようにデコーディングしているのかについての検討を行った。アレキサンダー病（AxD)はアストロサイト特異的分子GFAPの変異で起こる「一次性アストロサイト病」である。しかしミクログリアも本疾患に大きく影響していた。ミクログリアは、AxD病態を抑制する作用を有していることが明らかとなった。従ってよりミクログリアをより強力な細胞に置換することで、AxDの病態が大きく改善することが明らかとなった。また、ミクログリアのCa2+イメージングを可能とする遺伝子改変マウスを用い、AxD脳のミクログリアCa2+をin situで解析した。ミクログリアは、AxDアストロサイトの異常を感知し、Ca2+シグナル亢進としてその情報を読み出すことで、その表現型を大きく変化していることが明らかとなった。

Reason

次年度に予定していたAxD脳の一細胞シークエンスに成功し、その解析まで進んだこと、またミクログリアのin situ Ca2+イメージングにも成功し、AxD脳でミクログリアがどのようにAxDアストロサイトの情報を感知しているのかが明らかになったから、当初計画よりも進展していると判断した。

Strategy for Future Research Activity

一次性アストロサイト病であるアレキサンダー病（AxD）のモデルマウスを用い、ミクログリアがAxDアストロサイトの異常をどのように感知し、またどのようなメカニズムでその疾患に対応する応答を示すのか、の解析を行う。これまでに先行して行った一細胞シークエンスデータの解析、及び本年度開発したミクログリアのin situ Ca2+イメージングを平行して進める。AxD脳で大きく変化するミクログリアクラスターを同定し、これらの詳細な解析により、ミクログリアが如何にしてAxDアストロサイトの異常情報を感知し、応答しているのか（デコーディング）についての、分子メカニズムを明らかとする。
