Outline of Research at the Start

植物は、物理的接触や風雨による損傷などの過酷な物理的傷害ストレスに晒されても、柔軟なレジリエンスを発揮する。このため、傷害を受けた直後の植物は二次代謝を変化させ、生理防御応答を行う一方、数日以内に傷害組織の再構築を開始し、損傷・欠失した器官を再形成する。これら一連の傷害応答に対して、光や温度等の複合的な環境条件が大きく影響するが、植物が傷害情報とその後感知する環境情報を統御してレジリエンスを発揮するしくみは解明されていない。本研究では、傷害を受けた植物が不規則に変化する複合環境情報を統合し、数分から数日という時間軸で傷害応答を最適化する分子機構を解明する。

Outline of Annual Research Achievements

光情報が傷害修復応答を調節するしくみの解明を進めた。昨年度までの研究から植物が傷害に応答して茎葉を再生するためには転写因子を介した光シグナル経路の活性化が必要であることが分かっており、今年度はこの転写因子が直接制御する下流標的遺伝子の同定を進めた。これまでに取得したRNAseqデータからこの転写因子が根端メリステム形成を制御する遺伝子群の発現抑制に必要であることを見出していたため、EMSAとChIP解析によりこれらの遺伝子のプロモーター領域に直接結合するかどうかを検討した。一方、この転写因子は茎葉メリステム形成に関与する制御因子の発現誘導にも必要であることから、同様にEMSAとChIP解析を行った。また、RNAseqデータから傷害後に茎葉が再生する際にこの転写因子に依存して光合成関連因子の遺伝子発現が上昇することを見出したことから、この時期に光合成阻害剤を付与したところ、茎葉再生が著しく阻害された。また、光受容体に注目して茎葉再生に必要な光シグナル経路の同定を行った結果、青色光受容体のクリプトクローム(CRY)が茎葉再生に関与していることがわかったため、CRY変異体において遺伝子発現が変動する遺伝子としてオーキシン応答転写因子であるARFの解析を進めた。CRYとARFはアルファフォールドによる相互作用予測により特定のタンパク質ドメインが相互作用することが示唆されたた。そのため、表皮細胞におけるCRYとARFのBiFC実験およびタンパク質免疫沈降を行い、二つの因子が複合体を作成することを確認した。

Reason

今年度の結果から、光シグナルによって活性化される転写経路は根端メリステム形成の抑制、茎葉メリステム形成と光合成の促進という少なくとも3つの経路を直接制御することで傷害後の茎葉再生を促進することが明らかになった。また、傷害、高温ストレス付与後のHSFA1d遺伝子の転写開始点変化に注目し、その生物学的意義を検証する実験も行った。この転写開始点変化は5’UTRに検出され、高温条件下ではコントロール条件下より短い5’UTRが形成される。先行研究において5’UTRの長さがタンパク質の翻訳効率に影響するという報告があることから、long 5’UTRもしくはshort 5’UTRを含むHSFA1d-GFPベクターをシロイヌナズナやベンサミアナに一過的に導入して翻訳効率を調べたところ、short 5’UTRの方が高い翻訳効率を示すことが分かった。また、光受容体(CRY)と相互作用する因子の解析を進めた。CRY変異体において遺伝子発現が低下するオーキシン応答転写因子ARFを同定し、CRYとARFの相互作用をBiFC法およびタンパク質免疫沈降法により確認することができた。さらに、CRYと茎葉抑制ARF共通に制御される遺伝子群を同定することができ、CRYから出発してカルスからシュートを形成する分子パスウェイを明らかにすることができた。これらの成果は今年度に予定していた研究計画に沿ったものであり、本研究は概ね順調に進展していると判断される。

Strategy for Future Research Activity

これまでは変異体を用いた表現型解析から光シグナル経路が傷害修復に関与することを示してきたが、こうした遺伝学的手法では傷害前後のどの段階でこれらの経路が機能するのかを明らかにすることはできない。一方、傷害後の茎葉再生には光情報が必要であるのに対し、傷害を受ける前に植物が光を受けると茎葉再生は著しく抑制されることも見えてきており、いつ、どのように光シグナル経路が機能するのかをより丁寧な時系列で調べる必要があることが分かってきた。そこで今後はこれらの遺伝子をエストラディオール添加によってゲノム編集することのできる植物体を作成する。発芽後すぐから遺伝子機能を喪失する植物体、傷害直前、直後に機能欠損する植物体を作成し、これらの植物を用いて傷害修復能を解析する。またこうした光情報がクロマチン状態の変化を介して、長期的に傷害修復に影響を与えるという仮説を検証するため、光条件下、暗黒下で傷害を与えた植物組織片を用いてシングルセルレベルのマルチオーム解析を行う。光情報によってクロマチンのアクセシビリティと遺伝子発現が一細胞毎にどう変化するかを調べ、光照射のタイミングによってどのように異なった傷害応答が引き起こされるのかを明らかにする。また、ARFとCRYの複合体形成が証明できたので、次に複合体で発現が制御される遺伝子群を特定する。その遺伝子群の変異体解析を進め、カルスからのシュート再生の表現型を解析することで、CRYとARFの複合体から始まる分子カスケードを明らかにする。以上の解析から、光応答による器官再生メカメカニズムを明らかにしていく予定である。
