Outline of Research at the Start

植物の環境適応のための多層的な遺伝子発現制御において、エピゲノム制御が重要な役割を果たすと考えられるが、その分子メカニズムや意義は未だほとんど理解されていない。本研究では、不均一環境変動がもたらすヒストン修飾、DNAメチル化、ヌクレオソーム構造変化、高次クロマチン相互作用などの多層的エピゲノム変化や、それに伴って生じる転写開始点変化・スプライシング変化等が、環境応答遺伝子の発現制御と植物の環境適応に果たす役割を明らかにする。

Outline of Annual Research Achievements

佐瀬は、シロイヌナズナにおいて遺伝子転写産物が環境ストレスによってどのように多様化するのかについて解析を行った。また、エピゲノムが変化した変異体などの解析により、植物のエピゲノム制御によってトランスクリプトーム多様化がどのように引き起こされているかかが明らかになりつつある。現在まで にシロイヌナズナにおいてさまざまなエピゲノム変異体のトランスクリプトーム解析を行い、その成果を論文発表した。 稲垣は、ヒストンH3K4モノメチル化酵素群の制御機構の解析を進め、それらは転写共役的に働く酵素と転写非依存的に他のエピゲノム情報やゲノム情報を目印に働く酵素に大別されることを明らかにし、論文として公表した。さらに、H3K4ジメチル化を除去する脱メチル化酵素も転写共役的に働くことを示し、論文として公表した。

Reason

シロイヌナズナにおいて、エピゲノム修飾の変化により多くのmRNAの転写開始点やスプライシングパターンが変化することを明らかにし、その成果を論文発表した。

Strategy for Future Research Activity

エピゲノム変化がどのように転写開始点変化やスプライシングなどのRNAの転写後プロセッシングに影響するのかさらなる解析を進める。加えて、トランスクリプ トームが環境ストレス条件下でどのような転写制御を受けるのか、表現型への影響を含めた解析を行う。また、転写共役的なH3K4メチル化制御の意義を探るために、メチル化・脱メチル化酵素変異体を用いて、環境応答性遺伝子発現の解析を行う。
