In this research project, we aimed to investigate the diversity, generality, and mechanisms of recently emerged noncanonical translation dynamics, and obtained the following results. (Aim 1) Universality and molecular mechanism of ribosome instability dependent on newborn chains: After the discovery of the phenomenon in E. coli in which acidic residue-rich nascent chains destabilize translating ribosome (IRD), we found that a similar phenomenon can occur in eukaryotes, elucidated part of the molecular mechanism, and searched for factors that alleviate IRD. (Aim 2) We developed a method to search for new proteins that are not confined to known ORFs. (Aim 3) We investigated the molecular mechanisms and cellular dynamics of disease-associated noncanonical translation (RAN translation).
