Outline of Research at the Start

家族性筋萎縮性側索硬化症、脊髄小脳失調症など遺伝子ノンコーディングリピート配列の異常伸長を原因とする神経変性疾患において、リピート関連非AUG依存性（RAN）翻訳という新規の翻訳機構によりリピートペプチド（RAN-P）が産生され、神経変性が引き起こされる。本研究では、培養細胞やショウジョウバエモデルを用いて、RAN翻訳の分子メカニズム、そしてRAN-Pによる神経変性メカニズムを解明する。

Outline of Annual Research Achievements

遺伝子非翻訳領域内のリピート配列の異常伸長を原因とする筋萎縮性側索硬化症（ALS）、脊髄小脳失調症（SCA）などの神経変性疾患において、リピート関連非AUG依存性（RAN）翻訳という新規の翻訳機構によりリピートペプチド（RAN-P）が産生され、神経変性を引き起こすことが明らかになった。本研究では、RAN翻訳のメカニズムとそれに伴う神経変性メカニズムを解明することを目的として研究を行い、以下の成果を得た。
１）RAN翻訳レポーター細胞を用いたRAN翻訳分子機構の解明：C9-ALSの原因となるC9orf72遺伝子の異常伸長GGGGCCリピートの下流にNanolucを挿入したRAN翻訳レポーターコンストラクトを発現させた培養細胞発現実験系において、様々なRNA結合タンパク質（RBP）や翻訳開始因子、RNA輸送・分解関連分子などのノックダウンによるRAN翻訳に及ぼす影響を検討し、RAN翻訳活性に有意な変化を及ぼすいくつかの分子を同定した。
２）疾患モデル細胞・ショウジョウバエを用いたRAN翻訳の神経変性病態への役割の解明：異常伸長GGGGCCリピートRNAを発現するC9-ALSモデルショウジョウバエを用いて、様々なRBPや翻訳開始因子などの過剰発現／ノックダウンによるRAN翻訳制および複眼変性に対する影響を検討した。その結果、FUSはGGGGCCリピートRNAに結合してその高次構造を変化させてRAN翻訳を阻害し、C9-ALSモデルショウジョウバエの神経変性を抑制することを明らかにした。
３）RAN翻訳ペプチドによる神経変性機序の解明：RAN-Pを発現する培養細胞実験系において、RAN-Pが核小体に局在し、リボソームの合成を障害することを明らかにした。

Reason

RAN翻訳レポーターの再設計が必要となったために、研究計画を変更し細胞・ショウジョウバエ実験の開始が遅れたが、変更後は予定通り、研究計画を完了した。

Strategy for Future Research Activity

今後、以下のような研究を進める。
１）RAN翻訳レポーター細胞を用いた実験系を用いて、さらにRAN翻訳に影響を及ぼすRBPや翻訳開始因子、RNA輸送・分解関連分子などの同定を進めて、RAN翻訳のメカニズムを明らかにする。
２）疾患モデル細胞・ショウジョウバエを用いて、さらにRAN翻訳制御分子候補のRAN翻訳、細胞毒性および神経変性に対する影響を明らかにし、RAN翻訳の神経変性病態への役割を解明する。
３）RAN翻訳により産生されるRAN-Pが、RNA結合蛋白質などが液-液相分離介した液滴形成により形成するストレス顆粒など非膜性オルガネラの機能に与える影響を明らかにし、神経変性メカニズムを解明する。
