Outline of Research at the Start

Long non-coding RNA（lncRNA）の定義は「タンパク質をコードしない200塩基以上のRNA」とされているが、われわれはこれまでにlncRNAに存在する小さなORFから翻訳される新規タンパク質を同定した。すなわち、これらlncRNAは「ノンコーディング」ではなく「コーディング」RNAである。本申請課題ではこのような新規タンパク質を網羅的に同定し、生理機能を明らかにしていく。

Outline of Annual Research Achievements

Long non-coding RNA（lncRNA）の定義は「タンパク質をコードしない200塩基以上のRNA」とされているが、われわれはこれまでにlncRNAに存在する小さなORFから翻訳される新規タンパク質を同定し、それらが重要な機能を持つことを明らかにした [Matsumoto et al., Nature 541: 228-232 (2017)]。すなわち、これらlncRNAは「ノンコーディング」ではなく、「コーディング」RNAであり、存在すらしないと思われていた未開拓のタンパク質を翻訳している。lncRNAは進化に伴い数が増加し、組織特異的に発現するlncRNAはその数が進化と共に飛躍的に増加することが知られている。さらにlncRNAは、組織特異的(脳や精巣、血球系、肝臓など)に発現しているlncRNAが多い。すなわち、lncRNAと思われていたRNAから翻訳されるタンパク質も同様に、組織特異的に発現し、進化と共にその数が増加していくと考えられる。
これまでに新規ポリペプチドのリストを構築し、これら新規ポリペプチドについて複数のタグノックインマウスやノックアウトマウスを作成していた。そこで、ノックアウトマウスを用いて、行動解析や妊孕性の確認などを行い、さらにタグノックインマウスの組織を用いて、免疫沈降と質量分析解析により、新規タンパク質に対する結合タンパク質などを同定した。

Strategy for Future Research Activity

タグノックインマウスを用いた結合タンパク質のリストや、ノックアウトマウスのプレスクリーニングから得られた表現型の結果をもとにして、今後はさらに詳細な分子機構の解析などを進めていく。
