Outline of Research at the Start

染色体は、生命の設計図・ゲノムDNAの担体として、生命の活動と進化を支えている。細胞分裂期の染色体構築は、複製した遺伝情報を２つの娘細胞に分配するために必須な準備過程である。本研究では、この過程の主役である「コンデンシン」と呼ばれるタンパク質複合体が、どのような分子メカニズムを通して染色体構築を制御しているかという本質的な問題を明らかにする。

Outline of Annual Research Achievements

（１）染色体構築におけるループ押出し活性の貢献を明らかにする：コンデンシンIのループ押出しアッセイにおいてトポイソメラーゼIIを共存させたときに起こるDNAトポロジー変化の解析を開始した（領域代表・西山との共同研究）。興味深いことに、トポイソメラーゼIIはコンデンシンIによって形成されるループに結び目（knots）を導入し、よりコンパクトな構造に変換することを見出した。
（２）DNA超らせん化活性とループ押出し活性との関係を明らかにする：組換え型コンデンシンI複合体を用いてDNA超らせん化活性を測定する努力を続けているが、現時点ではいくつかの技術的問題に直面している。
（３）コンデンシン機能におけるHEATリピートと天然変性領域の役割を明らかにする：カエル卵抽出液を用いた実験により、CAP-D3 C-tailの欠失型変異（201アミノ酸欠失）がコンデンシンII による染色体構築を顕著に促進させることを見出し、論文として発表した。さらに、CAP-H N-tailの欠失型変異がコンデンシンI による染色体構築を促進するというデータも別報で発表することができた。

Current Status of Research Progress

Reason

（１）一分子アッセイを用いて、コンデンシンIとトポイソメラーゼIIの機能的協調作用を問うことに初めて挑戦し、興味深い結果を得ている。
（２）組換え型コンデンシンI複合体を用いてDNA超らせん化活性を検出することについては、さらなる条件検討が必要である。
（３）２つのコンデンシン複合体の天然変性領域による制御機構について２報の論文を発表できたことは高く評価したい。

Strategy for Future Research Activity

（１）染色体構築におけるループ押出し活性の貢献を明らかにする：コンデンシンIとトポイソメラーゼIIを組み合わせた一分子アッセイをさらに洗練することにより、これら２つの染色体構築タンパク質が一分子レベルでどのように協調して働くかを検討する。また、野生型と変異型コンデンシンIIのループ押出し活性を測定し、それぞれに対応するコンデンシンIのループ押出し活性と比較する。
（２）DNA超らせん化活性とループ押出し活性との関係を明らかにする：現在直面している技術的問題を解決することを優先させる。その後、Cdk1によるリン酸化反応の最適条件を見出すとともに、リン酸化がコンデンシンIの活性（DNA超らせん化活性とループ押出し活性）に与える影響を検討する。
（３）コンデンシン機能におけるHEATリピートと天然変性領域の役割を明らかにする：より短い領域を失った欠失型変異とより狭い領域の非リン酸化型変異を組み合わせることにより、CAP-D3 C-tailによるコンデンシンII活性の抑制メカニズムの詳細を検討す
