Outline of Research at the Start

ミクロンサイズの人工脳(ミニマル人工脳)構築に向けて,その基本単位となる人工細胞ユニット同士を連結させて動作する分子情報処理デバイスを設計・構築し,統合して働かせることを目指す.作製した分子デバイスは人工細胞膜上に存在し,特定の分子信号を認識して,状態を変化させ,膜越しに隣のユニット内部へとその変化を伝え,増幅するという機構を目指す.分子デバイスの設計は,人工ペプチド,人工核酸およびそのハイブリッドを用い,外部からの信号(光照射を予定)で状態をリセットし,再利用可能なものとする.これら人工細胞間の分子情報伝達機構を統合することで,他班と連携してミニマル人工脳の動作確認を目指す.

Outline of Annual Research Achievements

領域が目標に掲げるミクロンサイズの化学AIの構築に向け,本課題では,人工細胞膜小胞(GUV)の情報伝達を司り,かつ学習反応のためのリセットが可能な分子デバイスの設計と評価を行っている。本年度も引き続き,3種の情報伝達分子デバイス1)人工タイトジャンクション,2)人工レセプター,3)人工トランスデューサと,4)光照射によりリセット可能な分子デザインについて設計・構築と評価,検討を行った。野村グループは佐藤グループと共同で,引き続きDNAナノテクノロジーを基盤とした信号伝達分子デバイスの構築を行っている。GUVの膜上で機能する分子トランスデューサの設計と最適条件の探索を進め,核酸分子配列の伝達が直交性を有すること,さらにGUV内部のDNA回路を起動可能であることを示した。佐藤グループでは,さらに機能を高める分子トランスデューサとして,2つのGUV間における信号伝達機構を設計し,その動作確認に成功した。松浦グループは,昨年度のスピロピラン修飾光応答性ペプチドナノファイバーを用いた光による劇的なGUV変形の成功に続き,今年度は相分離GUVのLd相付近でペプチドナノファイバーを成長させることによりGUVの局所的な変形に成功した。今後、DNAシグナルに応答したGUV変形システムの創製を目指す。村山グループでは,人工核酸三重鎖で構成される膜貫通ドメインの合成スキームを改良し,合成期間の大幅な短縮を達成した。また,この膜貫通ドメインを介してGUV内のDNAと相互作用させることに成功した。これらの進展に加え,松浦グループはペプチド合成拠点を,村山グループは核酸合成拠点を引き続きそれぞれ運営しており,公募班を含む領域全体へのサービスと共同研究を引き続き強力に進めている。以上の成果に基づき,化学AI素子の実現のために他班・公募班との連携を進めている。

Reason

本年度も引き続き,3種+1項目の情報伝達分子デバイスについて設計・構築と評価を進めた。1)人工タイトジャンクション,2)人工レセプター,3)人工トランスデューサ,加えて4)上記の分子デバイスを再利用(リセット)可能にする機構,である。それぞれについて個々の進捗のみならず,共同研究による大きな課題へのアプローチが進んだ一年であった。下記のような特筆すべき進展があった。当初計画していたGUV間,隣接するGUVの内部から内部へ,脂質二分子膜2枚越しの情報伝達を実現する分子トランスデューサが,佐藤グループの設計によって確認された。これは,当初想定された機構よりも複雑化された,しかし昨年までに培った基礎的な知見に基づき実験上の課題を解決し得られた成果である。今回,設計された分子デバイスは世界にも類を見ない本領域発の新技術として発展的に用いられることが期待される。また,野村らはこれまでに発表した人工多細胞体(自己集合で形成されるcmスケールに及ぶヘミフュージョン体であり,各区画が脂質二分子膜で隔てられている)を簡便かつ大量に生産する手法を改良し,ロボットによる自動的な生成を実現し,報告した。この手法は本領域が目指す人工細胞間相互作用の構築と評価を行うための強力なプラットフォームとなり得る。さらに,松浦らは,スピロピラン修飾β-シート形成ペプチドによる光応答性可逆的なペプチドナノファイバーの形成・解離の制御に成功し,これを用いたGUVの大変形を,局所的に生じさせることに成功した。これらの進展は領域全体のデバイス統合による化学AI実現への課題解決を後押しする技術であり,今後の計画推進を強力にサポートする重要なものであると考えている。

Strategy for Future Research Activity

最終年度となる本年度は,これまで開発を続けてきた以下の情報伝達分子デバイスの設計・構築と評価,特に人工細胞膜および人工細胞間での機能評価に注力し,他班および公募班と連携してシステム化を追求する。人工タイトジャンクションとして選択的にGUV同士を隣り合わせて結合し,情報伝達を実現する機構と,人工レセプターとして,GUVの膜上に存在し,外部からの特定の分子信号と結合して構造を変化させ,溶液の交換を伴わずにリポソーム内部に状態変化を伝える機構,これらを統合して,2つの連結したリポソームの内部同士で状態変化を伝える機構とした(人工トランスデューサ)。これらのデバイスを実際にDNA回路を内包するGUVに組み込んで,連係した動作の評価を行う。さらに,外部からの光照射によるリセット機構,光応答性の局所的GUV膜の大変形機構を統合した一連のデザインの開発を進める.具体的な連携として,C01班,D01班で進行している学習・運動機構について,そのトリガーや,動作の結果を別のコンパートメントに伝える情報伝達について,我々の分子デバイスを活用した試験を行う。そして,A01班の開発している人工細胞回路の形成・処理を行う自動化機構と連携して,一連の分子情報伝達の評価を目指す。またペプチド合成拠点,人工核酸合成拠点による領域サービスの運営も引き続き行い,これら他班および公募班との連携を進め,分子サイバネティクス領域全体の成果としてのSPAユニットの動作を目指す。
