Outline of Research at the Start

学術変革領域「メガ生命深化動態」解析のモデルケースとして、バクテリアの内膜・外膜を舞台とするダイナミックなタンパク質輸送の基本原理の解明を目指します。本研究では関連するタンパク質の静的構造解析、動的構造解析などを統合し、タンパク質膜透過・膜組込み過程の分子機構の全貌を明らかとしていきます。将来的には、これらの解析方法を他の複雑な膜タンパク質解析系へと展開していきます。

Outline of Final Research Achievements

The dynamics of protein translocation across the membranes remain largely unknown. This study advanced the four-dimensional structural analysis of protein translocation across membranes. We prepared a single-unit protein translocation system using a nanodisc system and observed it by high-speed atomic force microscopy. We incorporated the latest AI technologies to analyze the images, revealing structural changes in the protein translocation motor during the protein translocation reaction.

Academic Significance and Societal Importance of the Research Achievements

解析のモデルケースとしてバクテリアの内膜・外膜を舞台とするダイナミックなタンパク質輸送を研究材料に、その基本原理の解明を目指しました。タンパク質の膜透過・膜組み込み過程は、生物に必須の細胞内機構の一つです。バクテリアでは内膜にSecトランスロコン(SecYEG)を中心とした複合体が存在し、Secトランスロコンをハブとするタンパク質輸送を担っています。この機構の解明は当該学術分野の最重要課題であり、異分野融合による精緻な解析を進めました。
