Outline of Research at the Start

超硫黄の生物学的意義を解明するには、システインパースルフィドなどの低分子超硫黄や、タンパク質中の超硫黄を定量的に解析する方法の確立が不可欠である。低分子超硫黄の定量的解析法はほぼ確立しつつあるが、タンパク質の超硫黄定量解析やプロテオーム、さらに、超硫黄分子イメージングにおいては、超硫黄側鎖の構造や機能解明の視点から、さらなる精度改良と世界標準化が望まれる。本研究では新規に開発した超硫黄プローブを用いて、定量的硫黄メタボローム・プロテオームのみならず、超硫黄オミックスイメージングを開発する。本領域で独自に確立する定量的超硫黄オミックスのプロトコールを国際標準化して国内外の超硫黄生物学を先導する。

Outline of Annual Research Achievements

本研究では、定量的超硫黄オミックスおよびイメージング技術の開発と世界標準化プロトコールとデータベースを構築する。この分析技術を基に、新たな疾病、老化制御・長寿医療の研究基盤を確立する。例えば、超硫黄オミックスやイメージングにより生体の硫黄代謝動態をモニタリングすることで、日常的な健康管理のみならず、動脈硬化などの生活習慣病、慢性炎症性肺疾患、心筋梗塞・心不全などの難治性心疾患などにおける病状把握や病態解明や予防・治療法の開発に繋げる。さらに、本超硫黄オミックス解析設備と技術を当該研究領域のバーチャルハブとして運用することで、総括班のミッションである領域内での横断的連携の強化のみならず、総括班にて先導する超硫黄のグローバルコンソーシアム構想と国際標準化・データベース化を支援し着実に遂行する。このことにより超硫黄生物学を世界的レベルで展開する。これまでに、超硫黄分子を安定的に捕捉するアルキル化プローブを用いた定量的超硫黄オミックス解析法を開発することにより、哺乳類・ヒトの細胞内に超硫黄分子が豊富に生成されることを見出した。加えて、単離ミトコンドリアを用いたシングル(単一)ミトコンドリアイメージングシステムを構築し、超硫黄分子を介したミトコンドリア膜電位形成を精度良く解析することで、真核生物における硫黄呼吸が、酸素呼吸とのハイブリット型システムであることを明らかにした。本年度では、環状八硫黄S8を特異的に捕捉する分子カプセルを用いた解析により、広く自然界に存在する環化S8が、生体内に高濃度で生成・蓄積していることを見出した。さらに、環化S8は、ミトコンドリアの硫黄依存型エネルギー代謝に関与する一方で、脂質過酸化を強力に制御することを明らかにした。引き続き、定量的な超硫黄分析技術を基にした超硫黄生物学研究を国際的に強力に推進する。

超硫黄分子の定量的オミックス・イメージング技術開発と標準化プロトコールの確立に向けた解析を行ったが、そのために本研究の当初目的の一つである、環化S8の定量解析法の開発とその生理機能解明に関する知見をさらに深めることができ、研究はおおむね順調に進展していると評価できる。

Strategy for Future Research Activity

本研究の当初目的に照らして、研究遂行の上での問題はなく順調に研究が進展している。今後は、定量的硫黄メタボローム・プロテオームのみならず、超硫黄オミックスイメージングの開発を推進する。さらに、本領域で独自に確立する定量的超硫黄オミックスのプロトコールを国際標準化して国内外の超硫黄生物学を先導する。
