Outline of Research at the Start

Gタンパク質には、7回膜貫通型受容体と共役する三量体Gタンパク質に加えて、低分子量Gタンパク質やダイナミン様Gタンパク質など、複数のファミリーが存在する。このうち、レドックス感受性の高い低分子量Gタンパク質H-Rasとミトコンドリア分裂促進Gタンパク質dynamin-related protein (Drp) 1の2つを用いて、タンパク質の超硫黄化の制御機構とシグナル変換/活性化のメカニズム解析を行い、心臓におけるその(病態)生理学的意義を明らかにする。これにより、タンパク質由来の高分子型スーパースルフィドの役割解明を目指す。

Outline of Annual Research Achievements

硫黄原子が複数連なったsulfane sulfur(超硫黄分子と呼ぶ)に対する選択的な蛍光指示薬SSip1, QS10などと、H2S/HS-に対して選択的な蛍光指示薬を組み合わせることで、心筋細胞や心筋組織におけるストレス誘発性の硫黄代謝・動態変化を可視化計測することに成功した。その結果、虚血や低酸素などのストレスによって心筋細胞内で超硫黄分子の異化が進み、それに伴って物理的な刺激に対する心筋の抵抗力が低下することが明らかとなった。その機序の一つとして、低酸素やタバコ煙曝露で誘導されるミトコンドリア過剰分裂に着目し、ミトコンドリア分裂促進Gタンパク質dynemin-related protein 1(Drp1)のC末端に存在するCys644残基が超硫黄化(Cys644-S(n)SH)されえていること、これが低酸素ストレス条件下で脱硫黄化されることにより活性化し、ミトコンドリア分裂が亢進することを明らかにした。さらに、Drp1のCys644残基は求核性が高く、そこに弱い親電子性を有する酸化型グルタチオン(GSSG)を処置することでCys644のグルタチオン化が生じ、かさ高くなることでDrp1活性を負に制御できることを新たに見出した。実際、心筋梗塞1週間後の心不全モデルマウスにGSSGと投与したところ、GSH投与しても改善されなかった心機能が、有意に回復することを確認した。以上の結果から、Drp1をはじめとするタンパク質Cysの超硫黄化が心臓のミトコンドリア品質や頑健性維持に関わっていること、GSSGが虚血性心不全に対する新たな治療薬となる可能性を示した。

申請者自身の主たる研究成果はNature Communication誌にリバイス対応中であるものの、国内外の他機関との共同研究成果がScience Advances誌やNature Communications誌に掲載され、一定の成果をあげることに貢献できている。タバコ煙の心筋ミトコンドリア毒性に関する研究成果や、心筋細胞・心線維芽細胞のリモデリングにおける超硫黄代謝の役割についての成果もJ. Pharmacol. Sci.誌に掲載されており、着実な成果を上げている。

Strategy for Future Research Activity

硫黄代謝を軸に心臓を構成する多細胞間の新たな相互作用や、心臓と他(多)臓器・組織との相互作用を明らかにすべく、心筋細胞や心線維芽細胞だけでなく、マクロファージや内皮細胞、肝細胞など様々な細胞種や組織を用いて超硫黄代謝の層別化・個別化の可能性を検証する。
