Outline of Research at the Start

アーキテクチュラルRNA (arcRNA) は、天然変性領域 (IDR) を持つ複数のタンパク質を集約して細胞内相分離を誘発し、非膜オルガネラの形成を主導する非ドメイン型RNAである。本研究では、近縁種間でのarcRNAの配列保存性の低さに注目して、一次配列以外の規則性に則って共通の機能を獲得するための非ドメイン型バイオポリマー特有の分子機構を解明する。さらにその機構に関わるarcRNA領域とIDRタンパク質が、非膜オルガネラの物性を決定している機構も取り上げ、arcRNAとIDRという二つの代表的な非ドメイン型バイオポリマーが協調して機能を獲得するための基盤則の確立を目指す。

Outline of Annual Research Achievements

1. 核内非膜オルガネラのパラスペックルは、核内空間で別の非膜オルガネラである核スペックルと隣接して存在しているが、決してお互いのオルガナイに取り込まれたり融合したりすることはなく、独立して存在している。このパラスペックルの独立性を規定する分子機構の解析を発展させ、これまで明らかにしていたパラスペックルと核スペックルの分離を促進する因子(SFPQ, HNRNPF, BRG1)に加えて、それと相反するパラスペックルを核スペックル内に取り込もうとする因子としてU2 snRNPを同定した。これによってNEAT1上の独立性を規定するRNA領域にSFPQら促進因子が結合することでU2の結合を妨げていることによって、パラスペッくるが核スペックルから独立して存在できることが示唆された。この独立性維持機構に関する論文は、Nature Cell Biologyに掲載され、その号のNews&Viewsに取り上げられるなど大きな反響があった。
2. 新たな非ドメイン型RNAとして、ストレスによって誘導される数百種類にも及ぶ3'伸長転写物DoGsを新たに同定した。これらのDoGsは、通常のRNA抽出に対して著しい難抽出性を示すことを見出し、そのために従来のRNA解析ではその存在が過小評価されていたことを示した。さらにDoGsがそれぞれ固有の核内非膜オルガネラに局在していることを見出し、arcRNAとしての機能がある可能性を見出し、この成果をRNA誌に報告した。さらにこれまでDoGsを誘導する浸透圧ストレス時に翻訳を阻害するという二重のストレス処理を実施すると、多くのDoGs誘導が著しく増加し、さらに難抽出性も著しく増加することを発見した。

Reason

これまで継続して実施してきたパラスペックルと核スペックルの独立性維持に関する成果を論文化し、一流ジャーナルに掲載することができたこと、さらに新たな非ドメイン型arcRNAとしてのDoGsが著しく誘導される条件を見出すことができ、このクラスのRNAの機能解析が大きく前進する可能性が見えてきたため

Strategy for Future Research Activity

非ドメイン型arcRNAの作用機構と生理機能に関する知見をさらに収集し、こうした分子種特有の機能獲得機構を明らかにするための実験を推進する。特に新たに多数同定されたDoGs RNAや難抽出性がゆえに見逃されていたarcRNAについての個別解析を進めて、非ドメイン型RNAの重要性を明確に提示するための一歩踏み込んだ知見を獲得する。さらに、こうした非ドメイン型arcRNAの探索法についてもタンパク質相互作用情報を活用した効率良い同定手順の確立を行い、さらなる非ドメイン型arcRNAの発見にこぎつけたいと考えている。
