Outline of Research at the Start

ほぼ全長に渡って構造をとらないと予測されるHeroタンパク質は、筋萎縮性側索硬化症 (ALS) の原因となるTDP-43などの「クライアント」タンパク質を様々なストレスから保護し、その変性や凝集体形成を強く阻害する活性を持つことが示されているが、その動作機構には大きな謎が残されている。そこで本計画研究では、Heroタンパク質によるクライアント保護機構の基本原理を解明すると共に、ヒト疾患iPS細胞ライブラリーを活用し、ヒト病態類似環境におけるHeroタンパク質の凝集体抑制効果を検証することで、新規の神経変性疾患治療法開発を目指す。

Outline of Annual Research Achievements

野生型TDP-43および家族性ALS患者に見られる病原性点変異を持つTDP-43変異体の一分子FRET観察を通じて、古典的分子シャペロンであるHsp40と、Heroタンパク質群の一つHero11が、ともに同様のTDP-43の構造変化を促し、より開いたコンフォーメーションを誘導することが確認された。この結論は、一分子FRETの詳細な時系列ダイナミクス解析によっても支持された。CRISPR-Cas9 sgRNAライブラリを用いた二段階のゲノムワイドスクリーニングで同定された、細胞機能の維持に必要な超天然変性タンパク質群に関しては、個々の因子の生理機能を探索し、いくつかの興味深い表現型が観察された。さらに、Heroタンパク質のヒト脊髄運動ニューロンにおける役割を評価するために、siRNAを利用してHeroタンパク質遺伝子のノックダウン(KD)実験を行い、ヒトニューロンおよびALS脊髄運動ニューロンでの発現変動を考慮し、15個の候補遺伝子を特定した。それらに対し、ノックダウンの条件を調整し、ニューロンの表現型に及ぼす影響を詳細に検討し、さらに絞り込みを進めた。また、CRISPR-Cas9システムを用いてΔNLS-TARDBP iPSCの作製に成功した。さらに、ALS疾患モデルであるQ331K変異を持つTDP-43トランスジェニックマウスの病態が、Hero11トランスジェニックマウスとの交配により改善されるかを検証し、細胞質に移行する型のTDP-43を表現するマウス(NEFT/TDPdNLS)を作成するため、必要な二つの系統を導入した。

一分子FRETイメージング、CRISPR-Cas9sgRNAライブラリーを利用した新規因子の探索、iPS細胞や培養神経細胞を用いた解析、モデルマウスを用いた解析、これらいずれも、上記概要の通り、おおむね当初の予定通り順調に研究が進捗していると判断できる。

Strategy for Future Research Activity

一分子FRETイメージング解析に関しては、これまでに得られた研究成果の総合的なとりまとめを行う。また、新たに同定された細胞機能維持に必要な超天然変性タンパク質群に対しては、個々の因子の生理機能の探求を進めると同時に、これらのタンパク質群の特性を包括的に分析し、体系的に理解を深める。これまでに開発されたヒト脊髄運動ニューロンの誘導方法、深層学習を利用した画像細胞解析システム、およびΔNLS-TARDBP iPSCモデルを組み合わせて使用し、健常者およびALS由来のiPSC運動ニューロンにHeroタンパク質の導入やノックダウンを行い、その影響を評価する。さらに、TARDBP変異モデルだけでなく、FUS変異モデルなど他のALS関連遺伝子変異を持つ運動ニューロンにおいても、Heroタンパク質の効果を検証する。また、Hero11の発現によってTDP-43(Q331K)マウスの組織学的な変化、すなわち神経変性や炎症反応の変化や改善があるかどうかを調べる。
