Outline of Research at the Start

極限環境耐性を持つクマムシは種特異的なタンパク質を多数有しており、それらの多くは煮沸処理後も可溶性を失わない「熱可溶性」という特異な性質を示す。本研究ではクマムシが示す優れたストレス耐性能力に注目し、クマムシ特異的な非ドメイン型タンパク質の細胞レベルでの機能解析を進めると共に、それらの分子レベルの作用機序から個体レベルでの生理機能まで、全階層横断的な解析を主導的に進める。さらに、比較ゲノム解析の手法を駆使して配列への機能依存性を高めずに機能を獲得する分子進化戦略を明らかにするほか、クマムシ特異的タンパクを強制発現し、新たな生体機能を持った個体の作製も目指す。

Outline of Annual Research Achievements

極限環境耐性を持つクマムシは種特異的なタンパク質を多数有しており、それらの多くは煮沸処理後も可溶性を失わない「熱可溶性」という特異な性質を示す。本研究ではクマムシが示す優れたストレス耐性能力に注目し、クマムシ特異的な非ドメイン型タンパク質の細胞レベルでの機能解析を進めると共に、それらの分子レベルの作用機序から個体レベルでの生理機能まで、全階層横断的な解析を主導的に進めることを目的としている。本年度は、これまでに開発したクマムシin vivo 遺伝子発現系TardiVecを用いたプロモーター領域の詳細解析により、CAHS3についてプロモーター活性を持つ領域を同定し論文発表した。また、Stereo-seqによる空間トランスクリプトーム解析によってクマムシにおける約16の細胞種の発現プロファイルを明らかにし、多くの細胞種で発現し、さらに緩歩動物門に広く保存された乾眠関連タンパク質を同定した。さらに、クマムシ類(緩歩動物門)で初めてとなるゲノム編集個体作出法の確立について論文発表するとともに、同技術を用いてノックアウト個体を作出することで、これまで機能未知であった新規なクマムシ非ドメインタンパク質について一部の極限環境耐性に機能的に寄与していることを明らかにした。同タンパク質はアミノ酸組成に著しい偏りがあり新規な耐性メカニズムの解明につながることが期待される。さらに、ゲノムリシーケンスとゲノム編集技術を組み合わせることで別途分離していたクマムシ変異体について原因遺伝子を同定できることを実証し、逆遺伝学に加えて順遺伝学的アプローチにも道を拓いた。また、非ドメイン型DNA防護タンパク質Dsupについて、生体内における流動性とそれに影響を与える領域を詳細に解析し、DsupによるDNA防護と生命活動の両立を説明する新しいモデルを提起した。

Reason

当初の予定通り、クマムシの乾眠に関わる非ドメイン型タンパクの機能解析と、新規タンパクの探索を進められている。予定通りTardiVecによるプロモーター配列の同定、空間トランスクリプトーム解析、CRISPR/Cas9系の確立やそれを用いたノックアウト実験を実行できている。

Strategy for Future Research Activity

予定通り比較ゲノム解析から明らかになった緩歩動物門に共通して保存される乾眠関連遺伝子についてそのクマムシ内発現に よるライブイメージングや機能解析を重点的に進める。これらの解析により、クマムシ乾眠における多数の非ドメイン型タンパクが組織・タイミング特性を持ちながらも協調的に個体の極限環境耐性を実現するレパートリーやメカニズムを明らかにすることを目指す。最終年度であるので、これまでの研究で論文になっていない部分を優先的にまとめていく。
