Outline of Research at the Start

上皮系幹細胞システムは、外界からのストレスや損傷に晒されながら個体を外界と隔て生命を維持している。申請者らは、哺乳類の成体表皮において個々の幹細胞同士が細胞競合を起こしてその恒常性を維持することを見出した。そこで様々なストレスに抗して幹細胞集団が自らを最適化し続ける自律性の原理は、生命を理解する上で根源的課題である。そこで様々なストレス下における基底膜分子の17型コラーゲンの発現の違いに注目し、マウス表皮幹細胞のシングルセルRNA-seq、多色クローン追跡、３次元培養表皮を組み合わせ、個々の上皮幹細胞がその違いに基づく競合的コミュニケーションを介して自律的に最適化してく仕組みを解明する。
