Outline of Research at the Start

本研究では、環境連成問題を徹底的に力学化して原生知能のアルゴリズムを読み解く。始めに、保存則を学理とした支配方程式を連立して解くことで、ジオラマ環境とパノラマ環境の物理量を詳細に求める。次に、細胞応答の数理モデル化を行い、個々の細胞の行動を計算する。最後に、細胞集団の行動を計算する「微生物行動シミュレータ」を開発する。開発したシミュレータを用い、密度成層内の有害赤潮藻の行動アルゴリズム、蠕動流れ中の腸内細菌の行動アルゴリズム、気液界面の微生物の行動アルゴリズムの3つの重要な生命現象を解明する。領域内の他班から実験データの提供を受け、自身は徹底力学化のインフラ的役割を担う。

Outline of Annual Research Achievements

本年度の主要な研究実績は以下の通りである。
1.繊毛が力を検知する機序を実験と数値シミュレーションによって調べた。開発した繊毛の計算力学モデルを用いることで、膜にかかる張力を定量的に評価した。この成果は、総合誌で世界最高峰のScience誌に掲載された(Katoh et al., Science)。 2.繊毛軸糸の詳細な構造と力学を再現した数理モデルを構築した。ダイニン分子モーターの活性を考慮することで、自律的に鞭毛打を生み出す機構を解明した。この成果は、生物物理学で定評のあるPhysical Review E誌に掲載された(Omori et al., Phys. Rev. E)。 3.ジオラマ行動力学の概念は、国際的にはまだ知れ渡っていない。この現状を克服するため、生物物理学で定評のあるBiophysical Journal誌において、ジオラマ環境の概念と行動力学の重要性について解説した(Ishikawa, Biophys. J.)。 4.多数の遊泳微生物の挙動を解析できるシミュレータを開発した。遊泳モードの異なる微生物のジェット流の挙動を調べたところ、プッシャー型とプラー型の両方とも不安定な挙動を示した。この成果は流体力学で定評のあるPhysical Review Fluids誌に発表された(Ishikawa et al., Phys. Rev. Fluids.)。 5.アメーバ運動を精度良く計測できる光学系を構築し、細胞膜の流動を計測することで、壁面上を前進するアメーバの力学モデルを構築した(Taniguchi et al., Biol. Open)。 6.ラッパムシが固着する際の周囲壁面の幾何学的な影響を調べた。その結果、ラッパムシが狭い場所を優位に選択して固着していることを発見した(Echigoya et al., Front. Cell Dev. Biol.)。

Reason

本年度は、鞭毛が力を感知する機序を解明した研究成果が総合誌で世界最高峰のScience誌に掲載された(Katoh et al., Science, 2023)。この成果は、NHK NEWS WEBやYahoo Japanニュース、Newsweek日本版、朝日新聞デジタルなどに掲載されて大きな反響を呼んだ。この論文のFWCIは47、Altmetricsは200と高く、Top 1%論文にカウントされている(Scopus調べ)。
採択後からこれまでに15編の査読付き雑誌論文と、27編の国際会議論文を発表しており、研究成果が勢いよく上がっていることから、当初の計画以上に進展していると判断した。

Strategy for Future Research Activity

2023年度は、微生物行動シミュレータの開発、個々の微生物行動の数理モデル化、微生物間の相互作用の数理モデル化と集団挙動の解析を行う。また、さまざまなジオラマ環境下における微生物行動を可視化計測し、新規行動アルゴリズムを探索する。
以下に具体的な計画を列挙する。
1)柴班や篠原班の実験で観察された精子の遊泳を3次元で再現できる微生物行動シミュレータを開発する。そして、背景流れや壁面境界、濃度勾配などの物理環境下における精子の行動を解明する。 2)中垣班が発見した「ラッパムシが狭い隙間を好んで付着する現象」に着目し、その行動アルゴリズムを議論する。狭い空間でラッパムシが作り出す流れを解析することで、ラッパムシが周囲の空間情報を認知する機序と、狭い領域に棲むことの利点を考察する。 3)紫加田班と共同して有害赤潮藻の挙動をマクロスケールで計測する。背景流れと光刺激が挙動に与える影響を体系的に整理し、それを再現できる微生物行動シミュレータを開発する。 4)繊毛が物理環境を感知するセンサーとして働く機構を調べるために、繊毛の計算力学モデルを構築する。そして、繊毛の屈曲や膜応力を解析し、力学検知のアルゴリズムを明らかにする。 5)微生物が凝集した環境下でどのように物質を輸送し、生命を維持しているのかを調べるためのシミュレータを開発する。それを用いたパラメトリックスタディを行い、その知見をより一般化するためのスケーリング解析も行う。 6)進化生物学のモデル生物であるボルボックス目に着目し、多細胞化することで得た高度なナビゲーションアルゴリズムを、バイオメカニクスの視点から明らかにする。 7)アメーバの細胞膜にマーカーを入れ込むことで、アメーバ運動時の内部および細胞膜の流動様式を可視化計測する。そして、仮足を伸ばして進行する行動力学を明らかにする。
