Germinal centers are the important site for the production of high affinity antibodies against pathogenic microbes. Our study revealed where in the lymphoid tissues B cells become committed to differentiation to germinal center B cells. Real time imaging analyses showed that the transcription factor Bcl6 plays an important role in B cell immigration into germinal centers. Imaging analyses also revealed that S1PR2, a receptor for sphingosine-1-phosophate, expressed by germinal center helper T cells is critical for their retention in germinal centers. In addition, our imaging analyses identified a subset of dendritic cells that is likely to be the most important for cytotoxic T lymphocyte development and differentiation, which are critical for tumor immunity.
