Outline of Research at the Start

申請者ら開発のCompton-PETは多分子の間接的な時空間相関イメージングが可能であるが、分子間相互作用を観測する医用診断装置は存在していない。
本研究では量子もつれ光子対放出原子核-分子間相互作用プローブおよびイメージング装置の開発を行い、次世代診断治療に必要となる薬剤分子間相互作用解析、悪性腫瘍・アルツハイマー病等の複数薬剤同時診断、個別化医療の実現が可能なイメージング治療技術基盤を創出することを目的とする。

Outline of Final Research Achievements

This study aimed to develop an unexplored diagnostic technique for detecting intermolecular interactions through precise measurement of cascade gamma photons correlated with nuclear spin. We successfully visualized pH-dependent anisotropy using a newly developed imaging system, demonstrating for the first time the potential to extract physicochemical parameters in vivo. By binding In-111 to a DOTA-based structure used for metal radiotracer delivery, we showed pH detection capability and confirmed its responsiveness in the pH 4-7 range. Intracellular uptake was also experimentally validated for the first time. Additionally, we demonstrated a twofold improvement in signal-to-background ratio using quantum entangled PET based on annihilation photon entanglement. Furthermore, we developed a detection method for local void size and oxygen concentration using the two-photon to three-photon ratio of positronium decay. These results confirm the success of our research objectives.

Academic Significance and Societal Importance of the Research Achievements

これまでは困難であった核医学領域における、量子センシング技術を新たに考案・開発し、pHやDOTA構造などの物理化学パラメータの抽出およびイメージングに初めて成功した。特に既存で用いられているSPECT核種を用いた、新たなイメージング技術の開発に成功したことは極めて大きな意義を有している。加えて、PET核種において、量子もつれ状態を用いたイメージング性能(信号背景比)の改善を、F-18のイメージング画像から初めて示した。さらに、新たなイメージング手法としてポジトロニウムの2光子/3光子比の利用を提案・可視化を実証した。今後医学診断技術に与える意義は大きい。
