Outline of Research at the Start

本研究では、放射性同位元素(RI)で標識した分子プローブを複数種作成し、生体内の異なる環境に導入してもつれγ線(光子)を測定することで、RIの種類・分子構造・生体内環境・もつれγ線の時空間相関の関係性を明らかにする。そして、生体・細胞・分子・原子・原子核にまたがる相互作用を理解し、最適なRIと分子の組み合わせを見つけることで、生体内の微小な化学的環境や分子間相互作用の変化をγ線の測定により非侵襲的に観測することを目指す。本研究の成果は、がんの治療薬の奏効率の判定等、患者さんごとに治療方法を最適化した個別化医療の実現につながると期待される。

Outline of Final Research Achievements

In this study, we aimed to observe the chemical environments inside cancer cells by using molecular probes labeled with radioisotopes (RIs) that emit entangled gamma rays, and by performing gamma-ray imaging capable of measuring perturbed angular correlation (PAC). We established a method for producing candidate isotopes for PAC spectroscopy, such as 155Tb, using an accelerator, and conducted preliminary gamma-ray imaging experiments. Meanwhile, we developed RI-labeled molecular probes targeting HER2 and created mouse models. We then performed in vivo entangled gamma-ray imaging experiments with Group A01. Although further optimization of experimental conditions is required to analyze the chemical environment in vivo through gamma-ray measurements, we obtained valuable data that will support future advancements in this field.

Academic Significance and Societal Importance of the Research Achievements

本研究の学術的意義の一つは、加速器を用いた155Tb等の大量製造法の確立に成功したことである。これらの核種は、本研究領域で開拓したもつれγ線イメージング法に限らず、医療用のSPECTイメージングや物性研究用の摂動角相関測定などにも使用でき、今後幅広い展開が見込まれる。一方、本研究では、RI標識分子プローブやマウスモデルの作成などを進め、生体内におけるもつれγ線イメージング実験を始めて実施することができた。今後、実験手法や条件を改善していくことで、がん細胞の位置と化学的環境を非侵襲的に測定できるようになり、革新的な医療診断技術を確立できると期待される。
