Outline of Research at the Start

免疫系細胞は極めて機能的多様性に富む細胞集団であり、恒常性維持や疾患の発症・増悪に深く寄与する。しかし、疾患時に様々な細胞が出現する場である間質に焦点を当てた統合的な研究は殆ど進んでいない。本研究計画では、病態の進行過程において対象臓器の間質に出現する免疫系細胞のサブタイプ(間質性免疫細胞とする)の解析を行うことにより、これまで支持組織として考えられていた間質側から新たに疾患を科学する。更に、シングルセル解析を応用し、病態に影響を与える間質性免疫細胞が選択的に相互作用する間質に存在している神経系、間葉系、血管・リンパ管や実質との複雑系細胞クロストークを解明する。

Outline of Final Research Achievements

In this study, building on prior findings that macrophages, once thought to be a single cell type, actually possess disease-specific subsets, we used the liver as a model to elucidate the dynamics and crosstalk of stromal immune cells during inflammation, fibrosis, and tumorigenesis. Single-cell RNA sequencing (scRNA-seq) analysis identified a novel myeloid cell population characterized by the expression of genes involved in angiogenesis. Furthermore, knockout of genes associated with this population significantly suppressed disease progression, suggesting a molecular basis by which stromal immune cells contribute to pathological tissue remodeling.

Academic Significance and Societal Importance of the Research Achievements

本研究は、疾患特異的なマクロファージサブセットの存在とその分子基盤を明らかにすることで、これまで不明であった間質性免疫細胞の多様性と機能的クロストークの実態に迫った点で学術的に大きな意義を持つ。特に、病態進行に関与する新規細胞集団とその標的遺伝子の同定は、線維化や肝がんといった難治性疾患に対する革新的な治療標的の創出につながる可能性がある。加えて、本研究で構築された間質セルアトラスは、疾患機序の再定義や個別化医療の基盤情報として社会的にも高い価値を有する。
