Outline of Research at the Start

実験技術の革新的な進歩により得られる大量のデータから生命現象の本質をどう抽出するか,さらには,複数の階層と時空間スケールからなる生命の構造をどう統合的に捉えられるかというデータと数理の問題は今後の生命科学の進歩を左右する極めて重要な問題の一つになっている.本研究では生命の各層から得られるデータから生命機能の本質的な情報を抽出し,それらの情報を階層間で繋げる.これにより生命現象全体を包括的に理解可能とする統合的数理手法の開発,及びデータから現象を結びつける実装的システムの基盤創出を目指す.

Outline of Annual Research Achievements

これまでの数理モデルを用いた生命現象の研究では,長年,現象論的なアプローチによってその本質を数学で記述し提案してきた.特に,複雑な生命現象を単純化する視点を生かした数理モデリングの研究は,生物実験だけでは発見することが難しい生命現象の本質を仮説として提案し,生物実験による検証を通じて「仮説」を「可能性」に変えることで,生命科学の発展に貢献してきた.一方で, 実験技術の革新的な進歩により,個々の細胞の動きや遺伝子の発現状態までを定量的に計測できるようになった.生命科学におけるデータ革命と言える時代の到来である.その一方,大量のデータから生命現象の本質をどう抽出するか,さらには,複数の階層と時空間スケールからなる生命の構造をどう統合的に捉えられるかというデータと数理の問題は未だ明確に解決しておらず,今後の生命科学の進歩を左右する極めて重要な問題の一つになっている.本研究では生命の各層から得られるデータから生命機能の本質的な情報を抽出し,それらの情報を階層間で繋げる.これにより生命現象全体を包括的に理解可能とする統合的数理手法の開発,及びデータから現象を結びつける実装的システムの基盤創出を目指す.

本年度では、新しいデータ記述法の開発を細胞とヒトの皮膚疾患において本格的に研究を進めた。まず、昨年度から進めていた細胞のlive imagingと数理モデルを統合する新しいモデリング手法の開発に成功し、その成果を論文としてまとめながら、国内外で積極的に発表を行った。さらに、ヒトの皮膚疾患においては、患者の皮疹の形状の情報から数理モデルのパラメータを推定できる全く新しい記述法の開発に成功した。

Reason

新しいデータ記述法となる細胞のlive imagingと数理モデルを統合する新しいモデリング手法の開発に成功し、論文を執筆中であり、近いうちに投稿予定である。これらの手法 はデータ科学と数理生物学の両分野において極めて斬新で新規性の高い手法であり、世界で初めての成果である。また、臨床医学に応用可能な新しい診断システムの基盤となる、皮疹の形状からのパラメータ推定システムの構築に成功した。現在、論文作成中であり、順調に研究が進展している。

Strategy for Future Research Activity

現象のデータを出発点とし,生命の階層で得られる実験データから情報を抽出できるデータ記述法を開発し,階層間のデータ情報を時空間で結びつけるデータ間の記述法を開発する.特に、データから数理モデル,そして数理モデルから現象をつなげるシステムを構築することで,実際の生命科学や医療現場でのデータを活用し,その本質的な仕組みを解明できるような記述手法を構築していく.これにより新薬や新しい治療法の開発に結びつける実装化の基盤を構築していく.以下ではそれぞれのサブプロジェクトにおける研究計画を述べる.
テーマ1)生命の多層にわたるパターン形成の統合的理解(GDA,TDA) 生命の各層のパターン形成をつなぐマルチスケールモデルの構築と,形と模様データの数学的定式化による実装基盤の構築:現在、新しいデータ記述法となる細胞のlive imagingと数理モデルを統合する新しいモデリング手法の開発に成功し、論文を執筆する段階に達している.本手法を実際のパターン形成と細胞形との関連性を解明する融合研究に応用していく.
テーマ2)細胞メカニクスに基づいた細胞機能操作の基盤創出(TDA) 流れ・形・パターンを統合的に考察可能な数理的手法の構築と細胞分化制御の非遺伝子操作手法の基盤創出:実験と3次元の細胞形状の数理モデルの融合により、細胞の変形による細胞内の物質の流れの現象を解明していく.
テーマ3)細胞内の遺伝子発現の情報から、細胞間の相互作用の抽出し数理モデリングに繋げる新しいモデリングアプローチを構築(PDA, GDA):単一細胞内の遺伝子の発現情報から細胞間のネットワークを推定し、細胞集団の動態の変化を記述する数理モデル手法の開発を行う。これにより、遺伝子レベルの情報から細胞、そして組織レベルまで統合できる数理モデルの構築に繋げる。
