Outline of Research at the Start

植物や微生物が作り出す『天然物』は人類社会を支えてきた。例えば、植物や菌類が生産するテルペン化合物は、医薬品・接着剤・電子材料などあらゆる分野に応用されている。これは、天然物が多種多様な構造と機能を提供してきたからに他ならない。言い換えれば、新奇天然物の発見は新材料・医薬品の開発に直結する。一方、天然からの新奇天然物の発見は年々困難となり、科学者に打開策が求められている。そこで本研究では、本領域の生合成実験科学者と綿密に協力しながら理論研究を推進し、天然物の巧みな『ものづくり』の仕組みを解明し、有機化学・天然物化学における学理・学術を提供するとともに、未踏天然物・非天然物の創出法を提供したい。

Outline of Annual Research Achievements

植物や微生物が作り出す『天然物』は人類社会を支えてきた。例えば、植物や菌類が生産するテルペン化合物は、医薬品・接着剤・電子材料などあらゆる分野に応用されている。これは、天然物が多種多様な構造と機能を提供してきたからに他ならない。言い換えれば、新奇天然物の発見は新材料・医薬品の開発に直結する。一方、天然からの新奇天然物の発見は年々困難となり、科学者に打開策が求められている。本研究では、理論計算によって得られた情報に基づき酵素・生合成経路を再設計・改変し、様々な実験グループとの共同研究を通して、天然には存在しない未踏天然物・非天然物の創出を目指している。本年度は引き続き、超連続反応である生合成経路解析のための理論解析手法の確立と、本手法を用いた生合成理論解析に取り組んだ。具体的には、いくつかのテルペン環化反応 などの酵素反応・生体反応について、反応経路の可視化に基づく、不安定中間体・遷移状態・軌道相互作用の系統的な探索を行い、Peniroquesine (JACS Au 2023)、 シクロプロパン環構築を触媒する生合成ラジカル反応メカニズム (J. Am. Chem. Soc. 2023)、脱炭酸転位反応を触媒する (Z)-Hinokiresinol 生合成メカニズム (J. Am. Chem. Soc. 2023)、シクロブタン環を足掛かりに立体選択的に官能基変換を行う生合成メカニズム (Angew. Chem. Int. Ed. 2023) などの未開目であった生合成経路/機構に関して、 既に全容解明に成功した。

Reason

本年度は学術変革研究の2年度目となり、高精度量子化学計算を用いた、複雑系生合成経路解析のための解析手法の確立を主な研究課題とした。解析手法の 確立は順調に進み、上記に示したように、本手法を用いた実際の生合成経路解析を行い、これまで未解明であったいくつかの生合成経路解析の全容解明に成功し化学系一般誌への論文発表も行っている。

Strategy for Future Research Activity

多種多様な天然物の骨格は、植物や微生物内に存在する種々の酵素内部での複雑な多段階連続反応によって、正確に、短時間で、効率的に生合成される。これら の酵素は、骨格構築酵素と修飾化酵素に分類することができる。これらの酵素群の機能を自在に設計・改変し、組み合わせることができれば、新たな骨格・構造 を持つ化学空間が開拓可能で、天然化合物ライブラリーの拡充が期待できる。そのためにも、酵素群の反応 制御機構の深い理解が必要である。 そこで、今後は 益々、本領域の生合成実験科学者と綿密に協力しながら理論研究を推進し、天然物の巧みな『ものづくり』の仕組みを解明し、有機化学・天然物化学における学 理・学術を提供するとともに、未踏天然物・非天然物の創出法を提供したい。
