Outline of Research at the Start

被子植物の遺伝的多様性を維持する生殖システムは、両性花を起点として多彩な進化を遂げており、本領域でも扱う性決定や自家不和合性などの他殖性機構を独立して種ごとに成立させるだけでなく、そのシステム間における自由な往来を可能としている。本研究では、両性花の活用を巡る幅広い植物群全体をカバーする横断的ゲノム解読によって、植物の進化全体を俯瞰する視点からゲノム・エピゲノム動態変化を検出する。各生殖システムの成立・破壊と連動するゲノム・遺伝子の進化パターンを数理的に定義するとともに、頻繁な生殖システムの移行に関わる潜在的な共通性を検証する。

Outline of Annual Research Achievements

被子植物の遺伝的多様性を維持する生殖システムは、両性花を起点として多彩な進化を遂げており、本領域でも扱う性決定や自家不和合性などの他殖性機構を独立して種ごとに成立させるだけでなく、そのシステム間における自由な往来を可能としている。本計画研究班では、両性花の活用を巡る幅広い植物群全体をカバーする横断的ゲノム解読によって、植物の進化全体を俯瞰する視点からゲノム・エピゲノム動態変化を検出する。各生殖システムの成立・破壊と連動するゲノム・遺伝子の進化パターンを数理的に定義するとともに、頻繁な生殖システムの移行に関わる潜在的な共通性を検証する。
染色体の端から端までを一続きで配列決定するテロメア・ツー・テロメア (T2T)全ゲノム解読を、クローバ、トマト、およびアセロラで達成し、植物ゲノムの完全解読への道筋を得た。植物のパンゲノム解析におけるT2Tゲノム解読の重要性を解説する総説を発表した。巨大なゲノムを持つ裸子植物(カラマツ、ヒノキ、スギ、およびコウヨウザン)ではゲノム解読結果を報告し、体細胞変異を利用することでクローンの系統解析が可能であることを示した。また、オオムギのパンゲノム解析により種内のゲノム変異を明らかにするとともに、イチジクの雌雄株のゲノム解析により性決定遺伝子領域を明らかにするなど、横断的ゲノム比較により種々の植物の多様性の変遷を明らかにした。領域内共同研究として、赤木班、藤井班、伊藤班、榊原班、奥田班、清水班、井澤班、須崎班と植物のゲノム解析を進めた。赤木班とはマンテマのゲノム解読を通して、植物の性染色体の進化を解明した。また、サクラやアサガオを題材としたアウトリーチ活動に取り組んだ。

Reason

両性花の活用を巡る幅広い植物群全体をカバーする横断的ゲノム解読を実施し、着実に成果を挙げている。ゲノム解読が及んでいない被子植物の基部群および複数の他殖性システムを有している目などの系統群を中心にしてロングリード技術やシングルセルゲノミクスの解析手法による高効率化を達成し、植物ゲノムの完全解読への道筋を得た。

Strategy for Future Research Activity

ゲノム解読が及んでいない被子植物の基部群および複数の他殖性システムを有している目などの系統群を中心にして、最新鋭のシークエンス技術を活用して、染色体レベルあるいはテロメア・ツー・テロメアレベルでのゲノム解読を行う。ゲノム構造、遺伝子配列、および転移因子等のリピート配列に着目して比較するための解析手法を確立する。
