Outline of Research at the Start

「多種多様な物質の識別」は免疫系が獲得した最も特徴的な能力である。この能力は、生物 が様々な外敵を感知するために進化してきたと考えられているが、近年、自己成分の認識にも関与していることがわかってきた。我々はこれまで、免疫受容体が認識する分子の同定プラットフォームを構築し、複数の自己リガンドを発見してきた。本計画研究では、この方法論を領域内で共有し、免疫センサーが認識する自己成分の戦略的同定を目指す。また、新たに同定した自己相互作用がもたらす生体応答の功罪 を明らかにすることで「免疫受容体による自己認識」が担う高解像度な自己監視機構の実態と免疫学を超えた普遍的な生物学的意義の解明を目的とする。

Outline of Annual Research Achievements

免疫系はこれまで非自己病原体を認識して排除する生体防御システムとして考えられてきた。しかし近年、免疫系が自己成分も認識していることがわかってきた。特に免疫系の自己認識には自己を攻撃する有害な「罪」の側面だけでなく、生体にとって有益な「功」の側面があることが明らかになりつつある。
我々のグループは、これまで免疫受容体が認識する生体分子の同定プラットフォームを構築し、C型レクチン受容体などの自然免疫受容体の内因性リガンドの同定や、MAIT細胞やNKT細胞などの自然免疫型T細胞の自己抗原の同定など、多くの自己反応性物質の単離・同定を行ってきた。本研究はこれまでに培った方法論を拡充し、自然免疫受容体が認識する自己成分の戦略的同定を目的としている。また免疫受容体の自己認識がもたらす「功」の側面を明らかにすることで、新たな生物学的意義を提唱し、免疫学に留まらない全生命現象の理解への貢献を目指している。
令和5年度は、昨年度から引き続き自然免疫受容体が認識する自己成分の同定を行った。その結果、自然免疫受容体の一つであり、真菌の受容体として知られていたDectin-1が、マウスの肝臓から抽出された水溶性画分を認識することを見出した。この水溶性画分は無菌マウスからも抽出されることから自己由来であることが考えられた。さらにDectin-1欠損マウスでは野生型マウスに比べて急性肝障害モデルや非アルコール性脂肪肝炎モデルにおいて炎症が改善していること、またDectin-1応答性の水溶性画分をマウスに注入すると肝炎を誘導することが明らかになった。
また、自然免疫型T細胞の制御に関わる自己由来脂質の構造活性相関研究、可視光応答性光触媒を用いた自己成分同定手法の開発基盤検討を行った。

Reason

本研究は免疫受容体の自己認識がもたらす生体保護作用を明らかにすることを目的としており、下記に示す研究課題を行うものである。
課題1:自然免疫受容体が認識する自己成分の戦略的同定
課題2:自然免疫型T細胞受容体が認識する自己保護性脂質の戦略的同定
まず課題1に関しては、他の領域メンバーと共同で、自然免疫受容体の網羅的なレポーター細胞ライブラリーの作成を進めている。またT細胞受容体の自己エピトープの同定に向けて内因性蛋白質由来ペプチド抗原ライブラリーを作成し、1段階のスクリーニングで簡便に抗原同定が可能となる検出系の確立を進めている。また課題2に関しては、自然免疫型T細胞受容体と抗原提示分子を導入したレポーター細胞を作成し、薄層クロマトグラフィーなどにより自己由来脂質を分離・精製し、レポーター細胞と共培養することで内因性リガンドの探索を進めている。
また、自然免疫型T細胞の制御に関わる自己脂質の構造活性相関研究を行ない、同定された自己由来の脂質関連化合物の各種誘導体を合成し、評価した。その結果、活性発現に関わる構造の一部を同定することに成功した。可視光応答性光触媒を用いた自己成分同定手法の開発基盤検討においては、ペプチド・タンパク質のタグ化を可能とする新たな反応形式の光触媒反応を開発した。自然免疫型T細胞の制御に関わる自己由来脂質の機能解析研究は順調に進んでいる。各種誘導体の合成ルートを確立し、合成・評価することで、当該脂質の構造活性相関情報を得ることに成功した。可視光応答性光触媒を用いた自己成分同定手法の開発検討についても、生理的条件下においてペプチドやタンパク質を修飾可能な独自手法を開発することができた。

Strategy for Future Research Activity

課題1に関する今後の方策としては、引き続き全ての自然免疫受容体を網羅したレポーター細胞ライブラリーおよび自己ペプチド抗原ライブラリーの構築を進めると共に、構築されたプラットフォームは計画・公募班員に広く提供し、自然免疫受容体が認識する自己成分の網羅的な同定を進める予定である。レポーター細胞で得られた活性画分はNMR測定や質量分析により化学構造の決定を行う。さらに化学構造が決定された場合には、自然免疫受容体のリガンド認識様式をクライオ電顕解析およびX線結晶構造解析によって明らかにする。
課題2に関しては、ヒトCD1aおよび拘束性T細胞受容体を発現したレポーター細胞を用いて免疫抑制効果を持つ自己由来脂質成分の検出を行う。抑制効果が検出された画分は、高速液体クロマトグラフィーや薄層クロマトグラフィーを用いて高純度に単離、精製を行い、NMR測定および質量分析によって化学構造の決定を目指す。活性脂質が同定された場合には炎症モデルマウスを用いて自己保護性脂質の抗炎症効果を評価する。
今後も前年度と引き続き、自然免疫型T細胞の制御に関わる脂質関連化合物の機能解析では自己成分同定手法の開発基盤検討を継続する。自然免疫型T細胞の制御に関わる自己脂質の同定・機能解析については前年度までに得られた構造活性相関情報を元にさらなる構造展開を実施し、活性発現に関わる鍵構造の同定を進める。また、NMRやLC-MSを用いた自然免疫型T細胞の制御分子の同定を継続する。可視光応答性光触媒を用いた自己成分同定手法の開発基盤検討では、前年度までに開発した手法について、細胞レベルにおいても化学修飾可能な手法へと展開するために、修飾タグや触媒の改変を検討する。
