Outline of Research at the Start

「多種多様な物質の識別」は免疫系が獲得した最も特徴的な能力である。この能力は、生物 が様々な外敵を感知するために進化してきたと考えられているが、近年、自己成分の認識にも関与していることがわかってきた。我々はこれまで、免疫受容体が認識する分子の同定プラットフォームを構築し、複数の自己リガンドを発見してきた。本計画研究では、この方法論を領域内で共有し、免疫センサーが認識する自己成分の戦略的同定を目指す。また、新たに同定した自己相互作用がもたらす生体応答の功罪 を明らかにすることで「免疫受容体による自己認識」が担う高解像度な自己監視機構の実態と免疫学を超えた普遍的な生物学的意義の解明を目的とする。

Outline of Annual Research Achievements

本研究では、生物有機化学的アプローチによって免疫センサーが認識する自己成分の同定・機能解析に取り組んでいる。当該年度は、前年度に引き続き(1)自然免疫型T細胞の制御に関わる自己由来脂質の構造展開、(2)可視光応答性光触媒を用いた自己成分同定手法の開発基盤検討を行った。
(1)自然免疫型T細胞の制御に関わる自己脂質の構造展開
山崎らによって同定された自己由来の脂質関連化合物の機能解析研究のために、新たな誘導体を設計合成した。
(2)可視光応答性光触媒を用いた自己成分同定手法の開発基盤検討
前年度までに開発した可視光応答性光触媒反応について、細胞上で反応を行うための条件を確立した。
本研究の学術的問いは、免疫受容体による自己認識が免疫系を超えてどのように生体に有益な応答をもたらすのか、また、その制御の破綻はどのようなリスクがあるのか、というものである。この問いに対して、免疫受容体の自己認識に関する進化的な研究はこれまでなされていなかった。これまでの免疫受容体に関する研究は主に哺乳類に限定されており、免疫受容体の進化的祖先が自己リガンドを認識するのか非自己リガンドを認識するのかが一切不明であった。そこで令和6年度は、自然免疫受容体の一種であるレクチン受容体に焦点を当て、ゲノムデータベースから進化的な起源となる遺伝子を探索・同定した。さらに、ゲノム解析で得られたレクチン受容体のリガンド認識に関する構造生物学的研究を行った。

Reason

自然免疫型T細胞の制御に関わる自己由来脂質の機能解析研究は順調に進んでいる。前年度に引き続き、機能解析のための新たな誘導体を設計し、合成ルートを確立した。
可視光応答性光触媒を用いた自己成分同定手法の開発検討についても、細胞上で反応を行うための検討を行い、条件を確立した。また、質量分析によって、タンパク質中のアスパラギン酸やグルタミン酸側鎖のカルボキシ基と特異的に化学修飾反応が進行することを確認した。さらに、光触媒からのエネルギー移動機構により化学修飾反応が進行することを確認した。
レクチン受容体遺伝子の脊椎動物における進化的保存性は不明であった。そこでヒトMincleに相同な遺伝子をデータベースから探索した所、硬骨魚類にも相同遺伝子が存在することを見出した。また、Mincleのアダプター分子であるFcRγ遺伝子も魚類で保存されていることを明らかにした。そこで魚類Mincleと魚類FcRγを培養細胞に共発現した所、協同的な発現の上昇を確認した。さらに魚類Mincleのリガンド結合ドメインの立体構造をX線結晶構造解析によって明らかにした所、魚類Mincleはリガンド結合ポケットが狭く、内因性糖脂質で見られる単糖は結合できるが、外因性糖脂質に見られる二糖以上の糖鎖は結合できないことが示唆された。

Strategy for Future Research Activity

今後も、引き続き、自然免疫型T細胞の制御に関わる脂質関連化合物の機能解析、自己成分同定手法の開発基盤検討を継続する。
(1)自然免疫型T細胞の制御に関わる脂質リガンドの構造展開、機能解析
自然免疫型T細胞の制御に関わる脂質リガンドの構造展開を進め、ケミカルプローブや創薬シーズとなる機能性脂質への展開を目指す。
(2)可視光応答性光触媒を用いた自己成分同定手法の開発基盤検討
化学修飾のさらなる効率化を図るため、修飾タグや触媒の改変を検討する。前年度に引き続きモデル細胞等を用いて、細胞表面分子の修飾検討を実施する。
これまでの研究から、レクチン受容体の祖先型遺伝子は自己認識に特化している可能性が示唆され、免疫受容体における自己認識機構が非自己の認識よりも普遍的な現象である可能性が考えられた。これは、免疫受容体は内因性分子の攪乱をモニターすることが普遍的な生理機能であることを示唆しており、自己認識機構を解明することの重要性を裏付けるものである。今後は、研究の対象を脊椎動物に拡大し、それぞれのリガンド認識特異性を明らかにすることで、外因性リガンドに対する認識能を獲得するに至った過程を調べる予定である。この研究によって、どのような選択圧がレクチン受容体の非自己の認識能獲得に至らしめたのかを明らかにしたい。
