Outline of Research at the Start

免疫系による自己成分の認識は、自己免疫疾患を引き起こすなど、生体にとって有害な反応であると考えられてきた。しかし、免疫系は本来、自己成分を認識するシステムであること、そしてそのシステムを通じて恒常性を維持していることが分かってきている。そのような自己認識機構を明らかにしようという学術変革領域研究の一つとして、本研究では、タンパク質の変動を感知するシステムおよびその破綻の分子基盤の解明を目指す。

Outline of Annual Research Achievements

本年度は、DNAリガーゼIV(LIG4)変異マウスの解析を進めた。獲得免疫不全と難治性炎症を示す患者において同定された、DNAリガーゼIVにアミノ酸置換(p.W447C)を引き起こす遺伝子バリアントをマウスに導入し、解析したところ、LIG4 W447Cホモ変異マウスにおいて、成長障害、DNA損傷修復障害と共に、B細胞、T細胞の顕著な減少(獲得免疫不全)が認められた。DNAリガーゼIVはDNA損傷修復の最終過程に必須の機能御分子であり、この表現型はDNAリガーゼIVの機能低下によるものと考えられた。
このLIG4 W447Cホモ変異マウスは、生後数か月以内に半数以上が、下痢、脱水など重篤な消化器症状を呈して死亡した。腸管、特に大腸では腸上皮の増生、杯細胞の消失と共に、炎症細胞の浸潤、陰窩膿瘍なども認められ、ヒト潰瘍性大腸炎様の病理所見が認められた。浸潤した炎症細胞に関しては、B細胞はほとんど認められず、CD4陽性T細胞(ヘルパーT細胞)とマクロファージが主体であった。また、炎症を来した腸管では、IFN-γおよびIFN-γで誘導される遺伝子群の発現が顕著に亢進していた。このような炎症病態は、放射線照射した野生型マウスに、LIG4 W447Cホモ変異マウス由来の骨髄細胞を移入したマウスでも認められた。以上の結果から、LIG4 W447Cホモ変異マウスにおいては、獲得免疫不全を背景にしながら、おそらく1型ヘルパーT細胞の活性化を基盤とした潰瘍性大腸炎様の病態が生じることが明らかになった。

Reason

獲得免疫不全下であるにもかかわらず獲得免疫の活性化により腸炎が生じる、新たな遺伝子改変マウスを作製することができた。これまでLIG4の低形成性変異マウスは2種類樹立されていて、いずれも成長障害、DNA損傷修復障害を呈し、悪性リンパ腫が発生することは報告されているが、炎症病態は報告されていない。また、リンパ球での遺伝子再構成を担う機能御分子RAGの低形成性変異マウスにおいて、獲得免疫不全下での獲得免疫の活性化、およびそれによる腸炎が報告されているが、それらのマウスではB細胞や、2型あるいは17型ヘルパーT細胞の活性化を示す点でLIG4 W447Cホモ変異マウスの表現型とは異なっている。このようなユニークな表現型を示す遺伝子改変マウスの樹立、解析は、自己指向性免疫学の功と罪のメカニズムの解明を目指す本研究に大きく貢献することが期待される。

Strategy for Future Research Activity

LIG4 W447Cホモ変異マウスに関して、T細胞を中心としたリンパ球、マクロファージを中心とした自然免疫担当細胞において、どのような異常が生じているのか、どのような細胞集団が病気の原因となっているのか解明を進める。IFN-γがどのような役割を果たしているのか明らかにする。またT細胞が関与している場合、T細胞受容体のレパートリー解析も行い、どのT細胞受容体が病気の原因となっているのか、解析を進める。
タンパク質の分解処理に異常を来す自己炎症性疾患モデルマウス、プロテアソーム変異マウスにおいては、好中球、単球においてどのような分化異常を来しているのか、どのいおうなタンパク質の蓄積によりその異常が生じているのか解析を進めると共に、適当な病態モデルを用いて、その病理的意義の解析も進める。さらに、B細胞、T細胞に生じている異常についても解析を進める。
タンパク質の細胞内輸送に異常を来す自己炎症性疾患モデルマウス、COPA変異マウスにおいては、マクロファージの異常、特に炎症性サイトカイン産生機構の異常について解析を進める。
