Research Abstract

1)極細胞形成因子であるミトコンドリアlrRNAの胚内分布
ミトコンドリア・ゲノムにコードされているlrRNAは、紫外線により失われた胚の極細胞形成能を回復せしめうることはすでに報告した。このミトコンドリアlrRNA(mtlrRNA)が、正常発生過程における極細胞形成においても機能していることを確認するために、in situ ハイブリダイゼイションを行った。
結果を要約すると:(1)産卵直後から卵割期にはmtlrRNAは卵後極の細胞質すなわち極細胞質に局在する、(2)核が後極に到達し極細胞形成が始まると、mtlrRNAは極細胞直下に極在し、極細胞中には入らない、(3)後極を紫外線照射した卵では、このmtlrRNAの局在が極めて微弱であるか、あるいは全く認められない。
さらに、電子顕微鏡レベルのin situハイブリダイゼイション技術を設立し、これを上記の胚に適用した結果、卵割期まではmtlrRNAは極顆粒上にのみ認められ、極細胞形成中に極顆粒から離れて細胞質中に遊離することが明らかになった。
さらにBicaudal-D突然異胚では、2次腹部が形成されるはずの前端部には局在していなかった。これは、mtlrRNAの極在が腹部形成ではなく極細胞形成に必要であることを示唆する。
2)卵形成過程における瀘胞細胞と生殖細胞との相互作用
エンハンサートラップ系より、卵の形成細胞巣中で、瀘胞細胞、卵細胞特異的に発現するものを選別し、これを卵形成ステイジのマーカーとして用いて、生殖細胞を欠く突然変異系統と、野生系との間で、これらマーカー遺伝子の発現状態を比較した。その結果、瀘胞細胞の分化には生殖細胞との相互作用が必要であることが明らかとなった。
