Research Abstract

今年度は、ショウジョウバエの複製因子であるDNAポリメラ-ゼ(以下polα)とPCNA(増殖細胞各抗原:polδ促進因子)の遺伝子の発現抑制機構の研究に重要な進展があった。
1)両遺伝子は共通の8塩基対TATCGATAの、促進配列(我々はDRE:DNA replicationーrelated elmentと命名した)によって支配されていることを証明したる。さらにこの配列に結合する分子量約85000のタンパク質因子DREFを発見し、その精製法を確立した。現在DREF遺伝子の単離を試みている。
2)昨年、両遺伝子の発現がホメオドメインタンパク質の一つ(zerknullt)によって抑制されることを報告した。この抑制と上記のDRE/DREFとの関係をしらべた結果、zerknulltはDREFの活性低下をもたらすことを証明した。この機構は細胞分化と共役した増殖停止にとって重要であると予想される。zerknulltがDREFを抑制する機構は不明であるが、DREFの遺伝子クロ-ニングおよびそれに対する抗体の作成が重要な手がかりを与えてくれるであろう。
